Se han analizado las propiedades coordinantes de estas prote\u00ednas con cd(ii) a ph 7 mediante valoraciones con dicho metal seguidas por varias t\u00e9cnicas: dicroismo circular, absorci\u00f3n electr\u00f3nica y espectrometr\u00eda de masas electrospray. El resultado de la valoraciones confirma la funcionalidad de las mt recombinantes y permite caracterizar por primera vez el fragmento beta de mt. Con el fin de aumentar la estabilidad de las proteinas en atm\u00f3sfera aer\u00f3bica se ha planteado la sustituci\u00f3n de residuos de cys por otros amino\u00e1cidos coordinantes y a la vez estables. El modelaje de formas mutantes de mt con cys substituidas por his, glu o asp ha predicho la viabilidad de la mutaci\u00f3n desde un punto de vista estructural. El estudio con p\u00e9ptidos sint\u00e9ticos derivados de mt permite comprobar que la sustituci\u00f3n de cys por his no impide que el p\u00e9ptidico coordine co(ii), zn(ii) o cd(ii). Finalmente se han estudiado las propiedades coordinantes de formas mutantes de mt obtenidas mediante t\u00e9cnicas de mutag\u00e9nesi dirigida. Se ha analizado como afecta la mutaci\u00f3n en la capacidad de unir zn(ii) in vivo i cd(ii) in vitro. El an\u00e1lisis de las propiedades \u00f3pticas y quirales de las prote\u00ednas ha permitido observar como afecta el cambio en la estructura de los agregados met\u00e1licos correspondientes y en la afinidad de la prote\u00edna por el cd(ii). El comportamiento de los mutantes demuestra que la mutaci\u00f3n no altera las habilidades quelatantes de la metalotione\u00edna.<\/p>\n<p>&nbsp;<\/p>\n<h3>Datos acad\u00e9micos de la tesis doctoral
