Coli del gen estructural vp60, y en el an\u00e1lisis de las principales regiones antig\u00e9nicas y inmunog\u00e9nicas de la prote\u00edna de c\u00e1pside del aislado de campo de rhdv, mc-89. La secuencia del gen vp60 present\u00f3 una heterogeneidad muy constante entre aislados co-circulantes (1-4,8%). Secuencias de virus m\u00e1s alejados como el ebhsv o el rcv permitieron identificar una regi\u00f3n con un nivel significativo de divergencia (3,45-44,4%). La producci\u00f3n en e. Coli de vp60 fusionada al ct de la beta-galactosidasa di\u00f3 lugar a prote\u00ednas de fusi\u00f3n que fueron r\u00e1pida y eficientemente proteolizadas por una diana tipo tripsina (r611-t612) presente en la beta-galactosidasa. La influencia del dominio heter\u00f3logo sobre el plegamiento de la beta-galactosidasa era reversible. El gen vp60 se subclon\u00f3 en cinco segmentos solapantes (aas 1667-1816, 1792-1940, 1924-2067, 2045-2198 y 2182-2344) y se expres\u00f3 en el vector pet-22b(+). En las prote\u00ednas recombinantes derivadas, la presencia de un p\u00e9ptido no hibrof\u00f3bico codificado por secuencias del mcs promov\u00eda la muerte celular y bajos niveles de acumulaci\u00f3n de prote\u00edna recombinante. La producci\u00f3n del segmento ct de la vp60 en dos vectores de la serie pet, revel\u00f3 que la versi\u00f3n citoplasm\u00e1tica se produc\u00eda en grandes cantidades, mientras que a presencia de un p\u00e9ptido se\u00f1al en la versi\u00f3n periplasm\u00e1tica de esta prote\u00edna generaba una disminuci\u00f3n dr\u00e1stica del crecimiento y muerte celular. En el estudio de antigenicidad (mediante western blot) de los segmentos recombinantes de vp60 se utilizaron sueros de animales vacunados e infectados con rhdv. Los resultados demostraron que, mientras el nt (158 primeros aas) estaba formado por determinantes antig\u00e9nicos con ep\u00edtopos b continuos, el ct estaba compuesto por ep\u00edtopos b dependientes de conformaci\u00f3n.
