Datos de nuestro grupo muestran que el tratamiento con dosis bajas de igf-i tiene efectos beneficiosos en la cirrosis experimental, mejorando la absorci\u00f3n intestinal de az\u00facares y amino\u00e1cidos, el balance nitrogenado, la osteopenia, y el hipogonadismo, adem\u00e1s de inducir un efecto hepatoprotector sobre el h\u00edgado cirr\u00f3tico. Sin embargo, todav\u00eda no se han explorado los mecanismos de este efecto beneficioso sobre la propia hepatopat\u00eda. Por otra parte, el principal mecanismo de hepatotoxicidad en la cirrosis inducida por cci4 es el da\u00f1o oxidativo por radicales libres que provoca peroxidaci\u00f3n lip\u00eddica, alteraci\u00f3n mitocondrial y da\u00f1o en el adn provocando la muerte celular, por necrosis o apoptosis. Con el fin de ahondar en los mecanismos del efecto hepatoprotector de igf-i sobre el h\u00edgado cirr\u00f3tico, los objetivos de este trabajo fueron: 1. Estudiar las pruebas de funci\u00f3n hep\u00e1tica, citolisis y colestasis en animales cirr\u00f3ticos con o sin tratamiento y en los correspondientes controles sanos; 2.  evaluar la lesi\u00f3n anatomopatol\u00f3gica; 3. Explorar la sensibilidad del h\u00edgado al factor de crecimiento determinando la expresi\u00f3n del receptor tipo i para igf-i; 4. Investigar la regeneraci\u00f3n hep\u00e1tica mediante el estudio por inmunohistoqu\u00edmica de la expresi\u00f3n del ant\u00edgeno nuclear de proliferaci\u00f3n celular (pcna) y mediante la cuantificaci\u00f3n de \u00e1cidos nucleicos, prote\u00ednas totales y contaje hepatocitario; 5. Estudiar la apoptosis mediante histoqu\u00edmica in situ (tunel) y la expresi\u00f3n de bcl-2 (prote\u00edna antiapopt\u00f3tica); 6. Analizar posibles mecanismos implicados en el efecto antioxidante y antiinflamatorio de igf-i en la cirrosis experimental, estudiando la expresi\u00f3n hep\u00e1tica de transferrina, inos, y tgf-beta1
