Socorro <\/strong><\/h2>\n<p>El presente trabajo de investigaci\u00f3n propone condiciones experimentales, de formulaci\u00f3n y preparaci\u00f3n, capaces de asociar la anfotericina b a dos pol\u00edmeros, poli(epsilon-caprolactona) por un lado, y poloxamer 188, por otro.  diversas t\u00e9cnicas, entre las cuales las espectroscop\u00edas de uv-visible, dicroismo circular y correlaci\u00f3n fot\u00f3nica, se han mostrado las m\u00e1s \u00fatiles, han permitido caracterizar de forma detallada la disposici\u00f3n y estado de agregaci\u00f3n de la anfotericina b en estos nuevos sistemas polim\u00e9ricos: la asociaci\u00f3n con nanoesferas de poli(epsilon-caprolactona) parece ser una adsorci\u00f3n en su superficie que disminuye la autoagregaci\u00f3n del f\u00e1rmaco.  la interacci\u00f3n con poloxamer implica la formaci\u00f3n de micelas mixtas que, contrariamente, aumentan el estado de agregaci\u00f3n de la anfotericina b. La afinidad del f\u00e1rmaco por ambos pol\u00edmeros es baja y, por tanto, se trata de asociaciones d\u00e9biles. En consecuencia, ambos tipos de sistemas son abiertos, o termodin\u00e1micamente inestables, y el f\u00e1rmaco se libera por simple diluci\u00f3n.  sin embargo, como ya se ha demostrado anteriormente, la vectorizaci\u00f3n en sistemas cerrados no es la \u00fanica estrategia para disminuir la toxicidad de la anfotericina b, y efectivamente, estos sistemas se mostraron capaces de disminuir la mortalidad que produce el f\u00e1rmaco in vitro, en eritrocitos, macr\u00f3fagos y c\u00e9lulas renales, as\u00ed como in vivo en ratones tras la administraci\u00f3n de un bolus intravenoso. Alteraciones en el estado de agregaci\u00f3n de la anfotericina b producidas por la elaboraci\u00f3n de estos nuevos sistemas, as\u00ed como el efecto de la presencia del poloxamer en la distribuci\u00f3n del f\u00e1rmaco desde ellos, podr\u00edan jugar un papel favorable en la disminuci\u00f3n observada en la toxicidad.  al mismo tiempo que estos sistemas disminuyen la toxicidad de la anfotericina b, aumentan su eficacia in vitro frente a candida albicans y leishmania donovani.
