{"id":38850,"date":"2018-03-09T09:38:37","date_gmt":"2018-03-09T09:38:37","guid":{"rendered":"https:\/\/www.deberes.net\/tesis\/sin-categoria\/estudio-de-procesos-de-plegamiento-desplegamiento-de-proteinas-mediante-calorimetria-cinetica\/"},"modified":"2018-03-09T09:38:37","modified_gmt":"2018-03-09T09:38:37","slug":"estudio-de-procesos-de-plegamiento-desplegamiento-de-proteinas-mediante-calorimetria-cinetica","status":"publish","type":"post","link":"https:\/\/www.deberes.net\/tesis\/quimica\/estudio-de-procesos-de-plegamiento-desplegamiento-de-proteinas-mediante-calorimetria-cinetica\/","title":{"rendered":"Estudio de procesos de plegamiento-desplegamiento de proteinas mediante calorimetria cinetica."},"content":{"rendered":"<h2>Tesis doctoral de <strong> Chakir Boulaich Mohammed <\/strong><\/h2>\n<p>1) se ha demostrado mediante el estudio de un sistema modelo apropiado (plegamiento-desplegamiento de ribonucleasa a ph \u00e1cido) que, tanto la calorimetr\u00eda isoterma de reacci\u00f3n como la calorimetr\u00eda de flujo, pueden utilizarse para estudiar la cin\u00e9tica de plegamiento-desplegamiento de prote\u00ednas.  2) con este prop\u00f3sito, resulta m\u00e1s conveniente, desde el punto de vista de tiempo y material, la calorimetr\u00eda isoterma de reacci\u00f3n.  3) se ha aplicado la calorimetr\u00eda de reacci\u00f3n cin\u00e9tica a la caracterizaci\u00f3n del plegamiento-desplegamiento de lisozima de clara de huevo en concentraciones comparativamente altas de cloruro de guanidinio, lo que nos ha permitido obtener valores de entalp\u00eda de desnaturalizaci\u00f3n a esas concentraciones altas de desnaturalizante, datos que ser\u00eda muy dif\u00edcil obtener de otra manera.  4) el disponer de valores de entalp\u00eda de desnaturalizaci\u00f3n para lisozima a altas concentraciones de guanidina ha permitido evaluar cin\u00e9ticamente la validez de los modelos de uni\u00f3n e intercambio, que se utilizan habitualmente para explicar el efecto de agentes desnaturalizantes sobre la estabilidad de prote\u00ednas. Se encuentra que, aun cuando ambos modelos son capaces de explicar cualitativamente los datos experimentales, desde un punto cuantitativo el acuerdo entre teor\u00eda y experimento es pobre. Se encuentra tambi\u00e9n que una simple relaci\u00f3n lineal entre entalp\u00eda de desnaturalizaci\u00f3n y actividad de guanidina proporciona una buena descripci\u00f3n fenomenol\u00f3gica de los
