Jes\u00fas <\/strong><\/h2>\n<p>La inestabilidad de microsat\u00e9lites debida a mutaciones en los genes reparadores de mismatch se ha implicado en la carcinog\u00e9nesis de diversas neoplasias. El an\u00e1lisis de p\u00e9rdidas de heterocigosidad permite identificar regiones cromos\u00f3micas implicadas en la carcinog\u00e9nesis. Los estudios sobre este fen\u00f3meno en los tumores del sistema nervioso son pocos y los resultados heterog\u00e9neos, en gran parte por la ausencia de criterios unificados para definir estas alteraciones.  objetivos: evaluar la aplicaci\u00f3n de un secuenciador autom\u00e1tico basado en tecnolog\u00eda fluorescente para el an\u00e1lisis de microsat\u00e9lites en c\u00e1ncer y aplicar esta metodolog\u00eda al estudio de tumores primarios del sistema nervioso.  material y metodos: los tumores estudiados fueron 56 gliomas, 32 meningiomas y 11 neurinomas, correspondientes a pacientes intervenidos en el complejo hospitalario universitario de santiago. Se trata de muestras preservadas e incluidas en parafina, de las que se seleccion\u00f3 una zona adecuada de tumor para la extracci\u00f3n del adn. El adn constitucional de cada paciente se extrajo a partir de sangre perif\u00e9rica. Hemos amplificado ocho loci microsat\u00e9lites (d12s391, d21s11, fibra\/fga, d19s394, d1s1656, d9s153, d9s1867, d6s1583), que posteriormente fueron analizados utilizando un secuenciador autom\u00e1tico alfexpress. El fenotipo de error de replicaci\u00f3n (rer+) se defini\u00f3 por la presencia de inestabilidad en al menos 2 de los 8 loci.  resultados: la identificaci\u00f3n de inestabilidad de microsat\u00e9lites y p\u00e9rdidas de heterocigosidad est\u00e1 dificultada por diversos artefactos comunes a todos los strs (bandas tartamudas, pull-ups, amplificaci\u00f3n preferencial y artefactos de la electroforesis) y por artefactos caracter\u00edsticos de cada microsat\u00e9lite (picos aislados o patrones de m\u00faltiples picos). El sistema fibra\/fga tuvo la mayor proporci\u00f3n de artefactos inespeci\u00edficos.
