Elena <\/strong><\/h2>\n<p>Hemos seleccionado 67 melanomas correspondientes a 48 pacientes, con el objeto de investigar la importancia de dos mecanismos oncogen\u00e9ticos en este tipo tumoral: la inactivaci\u00f3n de supresores tumorales y el fen\u00f3meno de inestabilidad de microsat\u00e9lites.  mediante pcr-sscps y secuenciaci\u00f3n directa estudiamos todo el gen cdkn2a\/p16 y los exones 4 a 8 del gen p53. Determinamos la expresi\u00f3n inmunohistoqu\u00edmica de las prote\u00ednas prb y p53. Amplificamos por pcr cuatro microsat\u00e9lites (th01, fes, vw y f13) en el adn tumoral y constitucional de los 48 pacientes, comparando ambas muestras en un sistema autom\u00e1tico de electroforesis (alf).  nuestros resultados fueron:  1. M\u00e1s de la mitad (59,09%) de los casos sobrexpresaban p53, sobrexpresi\u00f3n que correlacion\u00f3 con el fen\u00f3meno metast\u00e1sico aunque no con la progresi\u00f3n tumoral.  demostraron mutaciones en el gen en 3 casos, y en todos ellos la sobrexpresi\u00f3n de la prote\u00edna fue especialmente intensa y generalizada.  2. Un tercio (33,3%) de los melanomas perd\u00edan la expresi\u00f3n de prb, p\u00e9rdida que correlacionaba con la localizaci\u00f3n en \u00e1reas expuestas al sol pero no con la progresi\u00f3n tumoral.  3. Aunque un peque\u00f1o n\u00famero de casos mostraba alteraci\u00f3n simult\u00e1nea de prb y p53, la observaci\u00f3n m\u00e1s frecuente (61,4%) fue la alteraci\u00f3n de s\u00f3lo uno de ambos supresores, lo que sugiere una compensaci\u00f3n entre ambas v\u00edas de supresi\u00f3n tumoral.  4. S\u00f3lo caracterizamos una mutaci\u00f3n en cdkn2a y dado que se logr\u00f3 la amplificaci\u00f3n de al menos parte del gen en todos los casos, pensamos que su implicaci\u00f3n en el melanoma espor\u00e1dico es limitada.  5.
