{"id":39448,"date":"1999-01-01T00:00:00","date_gmt":"1999-01-01T00:00:00","guid":{"rendered":"https:\/\/www.deberes.net\/tesis\/sin-categoria\/caracterizacion-molecular-del-gen-oprl-de-pseudomonas-putida\/"},"modified":"1999-01-01T00:00:00","modified_gmt":"1999-01-01T00:00:00","slug":"caracterizacion-molecular-del-gen-oprl-de-pseudomonas-putida","status":"publish","type":"post","link":"https:\/\/www.deberes.net\/tesis\/quimica\/caracterizacion-molecular-del-gen-oprl-de-pseudomonas-putida\/","title":{"rendered":"Caracterizacion molecular del gen oprl de pseudomonas putida."},"content":{"rendered":"<h2>Tesis doctoral de <strong> Jos\u00e9 Juan Rodriguez Herva <\/strong><\/h2>\n<p>Las bacterias del g\u00e9nero pseudomonas poseen un alto inter\u00e9s biotecnol\u00f3gico por sus numerosas aplicaciones en el \u00e1mbito medioambiental, entre las que destacan su uso como biopesticidas, como agentes potenciadores del crecimiento vegetal, en el tratamiento in situ de zonas contaminadas, etc. el \u00e9xito de estos tratamientos, que suelen requerir la introducci\u00f3n de los microorganismos en el suelo o la rizosfera, depende en gran medida de la eficiencia con la que las bacterias colonicen dichos ecosistemas. De ah\u00ed la importancia de estudiar qu\u00e9 componentes de la envuelta celular est\u00e1n implicados en estos procesos de supervivencia y colonizaci\u00f3n. En este trabajo se ha determinado la funci\u00f3n celular del gen oprl de pseudomonas putida y su papel en el proceso de colonizaci\u00f3n del suelo y adhesi\u00f3n a semillas. Adem\u00e1s, tambi\u00e9n se ha estudiado el papel que juega el ant\u00edgeno o del lipopolisac\u00e1rido en estos procesos. el gen oprl codifica la lipoprote\u00edna asociada al peptidoglicano, perteneciente a la familia de prote\u00ednas pal de bacterias gram negativas. Este gen estaba localizado, en el cromosoma de p. Putida, dentro del grupo de genes tol, que codifican el denominado sistema tol-oprl de pseudomonas putida, un complejo proteico localizado en la envuelta celular e implicado en el mantenimiento de la integridad de la membrana externa. Las cepas portadoras de la mutaci\u00f3n oprl son m\u00e1s sensibles que su cepa parental a diversos compuestos (como detergentes, edta o algunos antibi\u00f3ticos), liberan enzimas peripl\u00e1smicas al medio extracelular y adem\u00e1s muestran defectos en los \u00faltimos estad\u00edos de la divisi\u00f3n celular. Sin embargo, oprl no es, como se postul\u00f3 en un principio, un gen esencial para la supervivencia de pseudomonas putida. En este trabajo, tambi\u00e9n se construy\u00f3 un mutante de p. Putida afectado en la expresi\u00f3n de la cadena lateral o del lipopolisac\u00e1rido y se estudi\u00f3 su capacidad para colonizar suelos no est\u00e9riles y<\/p>\n<p>&nbsp;<\/p>\n<h3>Datos
