Algunas cepas de rhizobium y bradyrhizobium poseen un sistema hidrogenasa que oxida el h2 generado en los nodulos y aumenta la eficiencia del proceso de fijacion de n2. La hidrogenasa de estas cepas es una proteina heterodimerica que contiene ni y grupos fe-s. En esta tesis se han estudiado la organizacion y funciones de los determinantes geneticos de la oxidacion h2 (genes hup) en rhizobium leguminosarum. Mediante secuenciacion de un fragmento de adn de 7,8 kb, analisis de la secuencia y caracterizacion fenotipica de mutantes con inserciones tn5, se identifico una agrupacion de ocho genes, designados hupijkhypabfcd, necesarios para la sintesis de una hidrogenasa activa. El producto del gen hupi es una rubredoxina y el de hupk probablemente una proteina de \u00abandamiaje\u00bb. Las proteinas hypabfd contienen motivos, cxxc, de union a metales y estan probablemente implicadas en la incorporacion de ni a la hidrogenasa. La proteina hypb posee multiples histidinas contiguas en el extremo n terminal y fue purificada a partir de su sobreexpresion en celulas de e. Coli por cromatografia de afinidad por ni. Mediante ensayos de dialisis de equilibrio se demostro que hypb se une en solucion a 4 iones ni2+ por monomero de hypb con una kd de 2,5 um.  utilizando anticuerpos contra hypb purificada se demostro la sintesis de hypb en celulas microaerobicas de r.  leguminosarum y en bacteroides de guisantes. Se propone que la proteina hypb une ni intracelularmente y esta implicada en su transferencia a la apoenzima durante el proceso de sintesis de la hidrogenasa.<\/p>\n<p>&nbsp;<\/p>\n<h3>Datos
