Mediante mutag\u00e9nesis dirigida por oligonucle\u00f3tidos se ha analizado la relevancia de estos puentes de hidr\u00f3geno en el mantenimiento de las propiedades estructurales, electr\u00f3nicas y funcionales de enzima.  mientras que el ion f\u00e9rrico del enzima salvaje es pentacoordinado y presenta un spin electr\u00f3nico alto, la variante asp235ala muestra tres especies espectrosc\u00f3picamente diferentes entre ph 5 y 9, todas ellas con un espectro electr\u00f3nico indicativo de hexacoordinaci\u00f3n: una especie de spin alto, que predomina a ph \u00e1cido y dos especies de spin bajo que se forman sucesivamente al aumentar el ph. El enzima variante muestra un perfil de actividad-ph en condiciones de estado estacionario pr\u00e1cticamente id\u00e9ntico al del enzima salvaje, si bien la actividad de la primera es tres \u00f3rdenes de magnitud inferior. Ello sugiere que el spin electr\u00f3nico de ion f\u00e9rrico del enzima no tiene una influencia determinante en la velocidad de oxidaci\u00f3n de citocromo c. La eliminaci\u00f3n del puente de hidr\u00f3geno entre el carboxilato de asp235 y his175, ligando proximal del ion f\u00e9rrico, resulta en un enlace de coordinaci\u00f3n m\u00e1s debil que, junto a los cambios que tienen lugar en el lado distal del hemo, permite explicar el incremento observado en el potencial de reducci\u00f3n del par fe3+\/fe2+ del enzima variante.  la variante trp51ala de ccp presenta tres especies espectrosc\u00f3picamente distinguibles en funci\u00f3n del ph, en las que el \u00e1tomo central de hierro es hexacoordinado. La especie dominante a ph \u00e1cido, ls1, posee un espectro electr\u00f3nico y de dicroismo circular magn\u00e9tico indicativo de spin electrbonico bajo y de una coordinaci\u00f3n axial bis-histidina.
