En esta tesis se han abordado dos objetivos: i) implicaciones de la regi\u00f3n terminal del m\u00f3dulo de uni\u00f3n a colina de la amidasa lyta de neumococo (motivos p5,p6 y 18 residuos terminales) en la organizaci\u00f3n estructural de la prote\u00edna y en el reconocimiento de colina y ii) implicaciones estructurales y funcionales de las diferencias en secuencia entre las amidasas de pared de neumococo codificadas por la bacteria y por sus bacteri\u00f3fagos. Para ello se ha procedido a estudiar cuatro mutantes de lyta obtenidos por truncamiento de la regi\u00f3n c-terminal (amidasas p6, p5, p4 y p5c) y 4 amidasas hom\u00f3logas a lyta, (lyta101, pa1, ej1 y hb1), codificadas por el neumococo at\u00edpico 101\/87 y los fagos dp-1, ej-1 y hb-3, respectivamente, caracterizando, en todos los casos, las diferencias en estructura secundaria, terciaria y cuaternaria, y en los equilibrios de uni\u00f3n a colina, producidas por la modificaci\u00f3n de la estructura primaria. las t\u00e9cnicas empleadas han sido dicro\u00edsmo circular, espectroscop\u00eda infrarroja con transdormada de fourier, calorimetr\u00eda diferencial de barrido y ultracentrifugaci\u00f3n anal\u00edtica.  a nivel de su estructura secundaria, todas las prote\u00ednas estudiadas son de tipo mixto y presentan un elevado contenido en hojas y giros beta, siendo peque\u00f1as las diferencias existentes respecto a la amidasa lyta. todas las mure\u00edn-hidrolasas caracterizadas hasta el momento presentan equilibrios de autoasociaci\u00f3n regulados por la uni\u00f3n de colina.
