La migraci\u00f3n leucocitaria hacia el foco inflamatorio tiene lugar a trav\u00e9s del denominado proceso cascada de adhesi\u00f3n leucocito-endotelio en la que participan mol\u00e9culas de adhesi\u00f3n, citoquinas y quimioquinas. La activaci\u00f3n del factor de transcripci\u00f3n nf-kb es un requisito necesario para la transcripci\u00f3n de las mol\u00e9culas mencionadas en respuesta a mediadores inflamatorios como tnf-alfa. El objetivo de la tesis fue analizar el efecto de agentes inhibidores de nk-kb (en concreto los agentes antioxidantes n-acetilciste\u00edna (nac) y pirrolind\u00ednditiocarbamato (pdtc) sobre la expresi\u00f3n de mol\u00e9culas endoteliales dependientes de nf-kb como las mol\u00e9culas de adhesi\u00f3n e-selectina, vcam-1 e icam-1 y las citoquinas il-6, il-8 y gm-csf. pdtc y nac inhiben la secreci\u00f3n, en respuesta a tnf-alfa, de las tres citoquinas en estudio. Dicha inhibici\u00f3n est\u00e1 mediada por el bloqueo de la activaci\u00f3n de nk-kb y consiguientemente de la transcripci\u00f3n de los genes que codifican para aquellas citoquinas. de forma similar, pdtc inhibe la inducci\u00f3n de la expresi\u00f3n, mediada por agentes inflamatorios, de e-selectina y vcam-1. Por el contrario, icam-1 no se inhibe en presencia de agentes antioxidantes. Esta falta de inhibici\u00f3n es debida al efecto inductor del agente antioxidante sobre la mol\u00e9cula icam-1.
