Cristina <\/strong><\/h2>\n<p>Se ha examinado la composici\u00f3n de subunidades e isozimas de la fosfofructokinasa (pfk) en c\u00e9lulas de tumor asc\u00edtico y gl\u00e1ndula mamaria murina, el tejido de donde procede el tumor. La pfk tumoral contiene predominantemente subunidades c, siendo las l m\u00e1s abundantes en gl\u00e1ndula mamaria, detect\u00e1ndose en ambos tipos de c\u00e9lulas una variante de la subunidad m de menor movilidad electrofor\u00e9tica. Hemos aislado la secuencia codificante completa de la pfk-c tumoral que corresponde a una prote\u00edna de 784 amino\u00e1cidos y 85,5 kda, en la que est\u00e1n conservados los residuos propuestos para la uni\u00f3n de substratos y efectores alost\u00e9ricos y cuya expresi\u00f3n se mantiene constante a lo largo del crecimiento del tumor en ratones, disminuyendo cuando cesa la proliferaci\u00f3n celular. Este enzima ha sido sobreexpresado en levadura habi\u00e9ndose caracterizado como un tetr\u00e1mero estable, cuyas propiedades reguladoras indican que la subunidad c es mayoritariamente responsable de la ausencia de activaci\u00f3n por fructosa-1,6-p2 y la inhibici\u00f3n por p-enolpiruvato, peculiares de la actividad pfk global en c\u00e9lulas tumorales.
