{"id":41732,"date":"1999-01-01T00:00:00","date_gmt":"1999-01-01T00:00:00","guid":{"rendered":"https:\/\/www.deberes.net\/tesis\/sin-categoria\/esteres-de-metabolitos-carboxilicos-agentes-insulinotropicos-in-vivo\/"},"modified":"1999-01-01T00:00:00","modified_gmt":"1999-01-01T00:00:00","slug":"esteres-de-metabolitos-carboxilicos-agentes-insulinotropicos-in-vivo","status":"publish","type":"post","link":"https:\/\/www.deberes.net\/tesis\/quimica\/esteres-de-metabolitos-carboxilicos-agentes-insulinotropicos-in-vivo\/","title":{"rendered":"Esteres de metabolitos carboxilicos: agentes insulinotropicos in vivo."},"content":{"rendered":"<h2>Tesis doctoral de <strong> Juan  Antonio Garcia Martinez <\/strong><\/h2>\n<p>En este trabajo se ha investigado, in vivo, el efecto insulinotr\u00f3pico de varios \u00e9steres de metabolitos carbox\u00edlicos, tales como los de los \u00e1cidos succ\u00ednico, glut\u00e1mico y pir\u00favico, debido a que son capaces de soslayar los defectos detectados en el transporte, forforilaci\u00f3n y en otros tramos del metabolismo de la d-glucosa en la c\u00e9lula beta pancre\u00e1tica, considerados responsables de la disminuci\u00f3n de la liberaci\u00f3n de insulina estimulada por glucosa, en algunos modelos animales de diabetes tipo 2. Los resultados de este estudio indican que los \u00e9steres de metabolitos carbox\u00edlicos act\u00faan como nutrientes secretagogs, in vivo, estimulando la secreci\u00f3n de insulina, tanto en la rata normal como en la diab\u00e9tica, sin ejercer acci\u00f3n glucagonotr\u00f3pica. Tambi\u00e9n se demuestra que algunos \u00e9steres del \u00e1cido succ\u00ednico son capaces, tras su administraci\u00f3n intraduodenal, de pasar al torrente circulatorio y, por lo tanto, de manifestar su potencial insulinotr\u00f3pico. Adem\u00e1s, se documenta que es posible dise\u00f1ar nuevos \u00e9steres del \u00e1cido succ\u00ednico, como el 1,2,3-tri(etilsuccinil)glicerol \u00e9ster, que gracias a su alto potencial insulinotr\u00f3pico evita la necesidad de suministrar dosis altas. Igualmente, la generaci\u00f3n de metanol, resultante de la hidr\u00f3lisis intracelular de los metil \u00e9steres, se puede evitar con el uso de \u00e9steres del \u00e1cido succ\u00ednico alternativos, que no s\u00f3lo siguen siendo potentes secretagogos de insulina, sino que, adem\u00e1s, algunos de ellos muestran una clara acci\u00f3n hipogluc\u00e9mica. por otro lado, todos los \u00e9steres de metabolitos carbox\u00edlicos estudiados in vivo, potencian la respuesta secretora de la c\u00e9lula beta a las sulfonilureas hipluc\u00e9micas y a los derivados de la meglitinida, y en algunos casos, tambi\u00e9n intensifican su acci\u00f3n hipogluc\u00e9mica. Todos lo documentado en este trabajo proporciona una serie de nuevos secretagogos de insulina, a considerar en la terapia de la diabetes tipo 2<\/p>\n<p>&nbsp;<\/p>\n<h3>Datos acad\u00e9micos de la
