{"id":42246,"date":"1999-01-01T00:00:00","date_gmt":"1999-01-01T00:00:00","guid":{"rendered":"https:\/\/www.deberes.net\/tesis\/sin-categoria\/mecanismos-moleculares-de-la-apoptosis-inducida-por-peptidos-inmunomoduladores\/"},"modified":"1999-01-01T00:00:00","modified_gmt":"1999-01-01T00:00:00","slug":"mecanismos-moleculares-de-la-apoptosis-inducida-por-peptidos-inmunomoduladores","status":"publish","type":"post","link":"https:\/\/www.deberes.net\/tesis\/quimica\/mecanismos-moleculares-de-la-apoptosis-inducida-por-peptidos-inmunomoduladores\/","title":{"rendered":"Mecanismos moleculares de la apoptosis inducida por peptidos inmunomoduladores."},"content":{"rendered":"<h2>Tesis doctoral de <strong> Juan  Antonio Oses Prieto <\/strong><\/h2>\n<p>Hemos demostrado que un p\u00e9ptido sint\u00e9tico (pa), que contiene un motivo estructural com\u00fan (motivo \u00ab2-6-11\u00bb) que se encuentra en diversas prote\u00ednas de la matriz extracelular y que es capaz de activar monocitos humanos en cultivo, induce apoptosis en los cultivos de monocitos, ya que tras la estimulaci\u00f3n de los monocitos con pa se produce un aumento del grado de condensaci\u00f3n de la cromatina y de la fragmentaci\u00f3n nuclear, as\u00ed como de la exposici\u00f3n de fosfatidilserina en la cara externa de la membrana plasm\u00e1tica y de la fragmentaci\u00f3n internucleosomal del dna. Adem\u00e1s, se observa proteolisis de dos biosubstratos de caspasas, parp y d4-gdi y una disminuci\u00f3n de los niveles de bcl-2.  los extractos nucleares de monocitos incubados en presencia del p\u00e9ptido pa, manifiestan una elevada actividad dnaasa con relaci\u00f3n a los extractos de monocitos no tratados. El aumento de actividad nucleasa que se detecta a ph 7 en presencia pero no en ausencia de ca2+ en los n\u00facleos de monocitos incubados con pa, disminuye por el pretratamiento de las c\u00e9lulas con el inhibidor de caspasas z-devd-fmk. estas nucleasas podr\u00edan estar reguladas tambi\u00e9n por poliadpribosilaci\u00f3n.  la apoptosis inducida por el p\u00e9ptido pa est\u00e1 mediada por las tirosina quinasas solubles src y syk, esfingomielinasa \u00e1cida y caspasas, as\u00ed como por p38. esta \u00faltima es activada por ceramida pero no por caspasas. La activaci\u00f3n secuencial de ceramida y caspasas lleva a la activaci\u00f3n de sapk.  este proceso est\u00e1 regulado positivamente por ampc, tal vez v\u00eda inhibici\u00f3n de pi3k. El no tendr\u00eda un papel antiapopt\u00f3tico.  hemos demostrado asimismo que la beta-endorfina induce tambi\u00e9n apoptosis v\u00eda ca2+ y camp.<\/p>\n<p>&nbsp;<\/p>\n<h3>Datos acad\u00e9micos de
