En este trabajo hemos investigado la eficiencia de los aav para transducir in vivo e in vitro c\u00e9lulas de carcinoma hepatocelular (hcc) y el uso de diferentes estrategias para aumentar la transducci\u00f3n de hcc. El pre-tratamiento de hcc con irradiaci\u00f3n gamma, con el inhibidor de la topoisomerasa denominado etop\u00f3sido o la infecci\u00f3n con adenovirus mejor\u00f3 significativamente la transducci\u00f3n de c\u00e9lulas in vitro. Esta mejora se debi\u00f3 a la conversi\u00f3n de la cadena simple del adn del aav a cadena doble en las c\u00e9lulas infectadas. en ratones inmunodeprimidos con tumores subcut\u00e1neos de hcc se observ\u00f3 la incapacidad de aav para transducir las c\u00e9lulas tumorales. Sin embargo, la administraci\u00f3n conjunta de aav junto con adenovirus result\u00f3 en una eficiente transducci\u00f3n en los lugares de la inyecci\u00f3n. La radiaci\u00f3n de los tumores subcut\u00e1neos con 1800 rad aument\u00f3 hasta un 30% la transducci\u00f3n mediada por aav. La infusi\u00f3n continuada de etop\u00f3sido a ratas buffalo con hcc dio lugar a la eficiente transducci\u00f3n de tumores m\u00e1s peque\u00f1os de 2 mm de di\u00e1metro. El tratamiento con etop\u00f3sido tambi\u00e9n aument\u00f3 la expresi\u00f3n del transgen en el h\u00edgado no tumoral. La determinaci\u00f3n de las concentraciones de etop\u00f3sido en tejido hep\u00e1tico revel\u00f3 una alta concentraci\u00f3n del f\u00e1rmaco en el h\u00edgado no tumoral y un casi indetectable nivel en tumores grandes. Nuestros resultados indican que la radioterapia no s\u00f3lo incrementa la transferencia g\u00e9nica mediada por aav en las c\u00e9lulas tumorales sino tambi\u00e9n aumenta la distribuci\u00f3n del vector en el interior del tumor. El etop\u00f3sido mejora la transducci\u00f3n de c\u00e9lulas de hcc in vitro y s\u00f3lo es eficaz in vivo en tumores muy peque\u00f1os.
