Otro punto importante, tambi\u00e9n encaminado a una aplicaci\u00f3n terap\u00e9utica es el estudio de la regulaci\u00f3n de receptores seroton\u00e9rgicos en determinadas patolog\u00edas del snc. Ante estas consideraciones nos planteamos dos objetivos claramente diferenciados: caracterizaci\u00f3n de posibles nuevos receptores 5-ht y regulaci\u00f3n de los subtipos 5-ht1a, 5-ht1b y 5-ht7.  para el primer objetivo se aplicaron dos t\u00e9cnicas de clonado por homolog\u00eda. La primera fue el rastreo de uangenoteca de cdna de hipocampo humano a baja astringencia empleando como sonda el gen codificador del receptor 5-ht1f de rat\u00f3n. Se lleg\u00f3 al aislamiento de 108 clones positivos, 5 de los cuales correspond\u00edan a subtipos de receptores seroton\u00e9rgicos de la familia 5-ht1 ya caracterizados. La segunda estrategia empleada fue la aplicaci\u00f3n de la pcr anidada para amplificar una genoteca de cdna de corteza frontal humana empleando como ampl\u00edmeros oligonucle\u00f3tidos degenerados dise\u00f1ados teniendo en cuenta los amino\u00e1cidos m\u00e1s conservados entre los receptores seroton\u00e9rgicos. se amplific\u00f3 un fragmento correspondiente al receptor 5-ht1a que presentaba una mutaci\u00f3n localizada al comienzo del segmento transmembrana iv. Esta mutaci\u00f3n hab\u00eda sido detectada por otros autores y parece corresponder a una variante de este receptor.  para el estudio de la regulaci\u00f3n de los subtipos de receptores seroton\u00e9rgicos se emplearon las neurotoxinas selectivas del sistema seroton\u00e9rgico para-cloroanfetamina (pca) y 3,4-metilendioximetamnfetamina (mdma o \u00e9xtasis). El efecto de las neurotoxinas se estudi\u00f3 tras administraci\u00f3n aguda y cr\u00f3nica a ratas y en cultivos primarios de embriones de rata.
