Estudios previos demostraron que la estimulaci\u00f3n a trav\u00e9s de cd50 induc\u00eda la movilizaci\u00f3n de calcio, incrementaba la funci\u00f3n beta1 y beta2 integrina y pose\u00eda efectos coestimuladores de la activaci\u00f3n t. El objetivo de este estudio ha sido establecer la relaci\u00f3n estructura\/funci\u00f3n de la regi\u00f3n citoplasm\u00e1tica de cd50. Con este fin, se han dise\u00f1ado diversas construcciones moleculares de cd50 basadas en mutaciones a nivel de la regi\u00f3n citoplasm\u00e1tica (formas truncadas o formas con substituciones aminoac\u00eddicas puntuales de s por a o y por f) que posteriormente se han transfectado a una clona deficiente para cd50 (camy.2) derivada de jurkat. Se han identificado dos regiones diferenciadas a nivel de la regi\u00f3n citoplasm\u00e1tica implicadas en la movilizaci\u00f3n de calcio a trav\u00e9s de cd50. Por un lado, una regi\u00f3n localizada en la parte m\u00e1s distal de la mol\u00e9cula, en la cual se ha localizado concretamente el residuo s515 como un residuo clave en la capacidad de movilizar calcio dependiente de cd50. Por otro lado, la regi\u00f3n correspondiente al tercio m\u00e1s proximal a la membrana de la cola, d\u00f3nde los residuos clave identificados con la s487 y la y490. El estudio del patr\u00f3n de fosforilaci\u00f3n de cd50 demuestra que se encuentra basalmente fosforilado a nivel del tercio m\u00e1s distal de la cola citoplasm\u00e1tica, mientras que la fosforilaci\u00f3n inducible de la mol\u00e9cula se localiza en el tercio m\u00e1s proximal a la membrana.
