{"id":4441,"date":"1994-01-01T00:00:00","date_gmt":"1994-01-01T00:00:00","guid":{"rendered":"https:\/\/www.deberes.net\/tesis\/1994\/01\/01\/diseno-y-sintesis-de-peptidos-inhibidores-de-la-infectividad-del-virus-del-sida-vih\/"},"modified":"1994-01-01T00:00:00","modified_gmt":"1994-01-01T00:00:00","slug":"diseno-y-sintesis-de-peptidos-inhibidores-de-la-infectividad-del-virus-del-sida-vih","status":"publish","type":"post","link":"https:\/\/www.deberes.net\/tesis\/quimica\/diseno-y-sintesis-de-peptidos-inhibidores-de-la-infectividad-del-virus-del-sida-vih\/","title":{"rendered":"Dise\u00f1o y sintesis de peptidos inhibidores de la infectividad del virus del sida (vih)."},"content":{"rendered":"<h2>Tesis doctoral de <strong> Isidro Prieto Santiago <\/strong><\/h2>\n<p>Se sintetizaron tres grupos de peptidos para su uso en la inhibicion de la infectividad del virus del sida (vih):  (1) pentadecapeptidos solapados de la glicoproteina de envuelta viral (gpl20), utilizados para obtener anticuerpos frente a los mismos en ratones balb\/c, mediante inmunizacion conjunta con el determinante t helper fiseaiihvlhsr; (2) derivados de la secuencia pol(151-161), como posibles inhibidores de la proteasa viral; y (3) derivados de cd4(81-92), con determinados residuos sustituidos por fenilalanina, como posibles inhibidores de la interaccion gpl20-cd4. Se obtuvieron anticuerpos frente a casi todos los peptidos de gpl20.  asimismo, casi todos los peptidos fueron reconocidos por uno o mas sueros procedentes de pacientes vih-1-seropositivos. La presencia de muchos de estos anticuerpos en los pacientes, postulamos que podria deberse a su elicitacion por fragmentos de gpl20. Los anticuerpos obtenidos mediante inmunizacion conjunta con fiseaiihvlhsr de los peptidos 61-75, 86-100 106-120, 206-220, 301-315, 306-320, 421-435, 471-485, 481-495, 486-500, 491-505, 496-510 y 491-505 reconocieron a gpl20 recombinante. De estos antisueros solo mostraron cierta actividad neutralizante los obtenidos frente a 106-120, 301-315, 306-320, 486-500 y 491-505.  los peptidos pl56c, pl56w y pl56y derivados de la secuencia pol(151-161) son inhibidores de la infeccion por vih-1 in vitro, con ic50s de 0,18; 4,33 y 15,35 m-respectivamente en celulas molt-3, y de 13,64; 3,44 y 12,53 m respectivamente, en celulas ht4lacz-1.  los mejores inhibidores de la infeccion in vitro en celulas ht4lacz-1 obtenidos a partir de la secuencia cd4(81-92) fueron fyicfvedokeedd (ic50= 2,20 m), fyicfvede (ic50= 1,60 m) y fyicfved (ic50= 1,22 m).
