Paz <\/strong><\/h2>\n<p>El tp53 es el gen mas frecuentemente mutado en tumores humanos. El objetivo de este estudio es valorar la participacion del gen tp53 en astrocitomas fibrilares difusos de adultos y en gliomas pediatricos del tronco cerebral. Se analizaron 34 astrocitomas difusos de adultos mediante analisis de polimorfismos en la conformacion de dna monocatenario y secuenciacion directa para deteccion y caracterizacion de mutaciones en los exones 5 a 8 del gen tp53; ademas se realizo inmunohistoquimica con el anticuerpo pab1801 para deteccion de la proteina. Trece de los 34 astrocitomas no mostraron alteraciones ni en el gen p53 ni en la proteina; 11 presentaron mutaciones en el gen, que daban lugar a proteinas alteradas; y 10 presentaron acumulacion de proteina, sin detectarse mutaciones en el gen. Este ultimo grupo no mostro alteraciones cuando se estudiaron los exones 4, 9 y 10 del gen p53 y solo uno de ellos reacciono con el anticuerpo pab240; ademas, ni amplificacion del gen mdm2. En 7 gliomas pediatricos del tronco cerebral se estudiaron perdidas de heterocigosidad en los cromosomas 10 y 17, y amplificacion del gen del receptor del factor de crecimiento epitelial mediante pcr diferencial. Se encontraron perdidas frecuentes en el brazo corto del cromosoma 17 y mutaciones en el gen p53, sin amplificacion del egfr, con perdida parcial del cromosoma 10 en algunos casos.
