En condiciones basales glut1 y los receptores de insulina se distribuyen mayoritariamente en membrana de superficie. La mayor parte de glut4 es intracelular. Glut1 es exclusivo de sarcolema y los receptores de insulina se localizan en tubulo-t y dominios selectivos de sarcolema. La insulina promueve la translocacion de glut4 desde una poblacion de vesiculas intracelulares a tubulo-t y a dominios selectivos de sarcolema. En respuesta a la insulina se produce la internalizacion de receptores de insulina a membranas intracelulares. Se han identificado 3 poblaciones de vesiculas intracelulares que contienen glut4 en musculo esqueletico: una poblacion sensible a la insulina (participa en la translocacion de glut4), una insensible a la insulina y una poblacion que se enriquece en receptores de insulina en respuesta a la hormona. Ni las proteinas scamps ni caveolina parecen estar implicadas en la via de trafico intracelular de glut4. En cardiomiocitos de rata la insulina promueve la translocacion de glut1 y glut4. En este tipo celular existe un mayor grado de colocalizacion intracelular entre glut4 y proteinas scamps.
