Vrk2 end\u00f3gena se localiza en el n\u00facleo y en el citosol, como consecuencia de la expresi\u00f3n de dos isoformas generadas mediante un procesamiento alternativo del mensajero de vrk2, que da lugar a prote\u00ednas de 508 y 397 amino\u00e1cidos. Vrk2a contiene una regi\u00f3n hidrof\u00f3bica que ancla la prote\u00edna a la membrana del ret\u00edculo endoplasm\u00e1tico y mitocondrias y se expresa en la mayor\u00eda de tipos celulares estudiadas, mientras que vrk2b se localiza libre en el n\u00facleo y en el citosol y se expresa en l\u00edneas celulares en las que vrk1 es citos\u00f3lica. Ambas isoformas comparten un dominio catal\u00edtico id\u00e9ntico y fosforilan p53 in vitro \u00fanicamente en treonina 18. Solamente la sobre-expresi\u00f3n de la isoforma nuclear vrk2b induce la estabilizaci\u00f3n de p53 mediante un mecanismo postraduccional, principalmente debido a la fosforilaci\u00f3n en treonina 18, que induce la disminuci\u00f3n de la ubiquitinaci\u00f3n por mdm2 y promueve la acetilaci\u00f3n por p300. En este contexto, vrk2b podr\u00eda reemplazar funcionalmente a la quinasa nuclear vrk1, formando parte de rutas de se\u00f1alizaci\u00f3n implicadas en procesos proliferativos. la prote\u00edna de anclaje jip1 ensambla complejos de se\u00f1alizaci\u00f3n compuestos por mapk quinasas. La actividad de \u00e9stos es modulada mediante interacciones con otras prote\u00ednas. Vrk2 co-localiza con jip1 en la regi\u00f3n perinuclear, e interacciona de forma estable con el dominio c-terminal de jip1. No obstante esta asociaci\u00f3n no interfiere con la interacci\u00f3n entre jip1 y tak1, mkk7?1 o jnk. Sin embargo, vrk2 disminuye la activaci\u00f3n de jnk detectada como una reducci\u00f3n en su fosforilaci\u00f3n y la disminuci\u00f3n de la activaci\u00f3n de la trasncripci\u00f3n dependiente de ap1. De esta forma vrk2 modula la respuesta celular inducida por interleuquina-1?
