En la ruta biosintetica del peptidoglicano est\u00e1n involucrados 3 compartimentos celulares: el citoplasma, la membrana citiplasmatica y el periplasma. La \u00faltima fase de la biosintesis del peptidolgicano es llevada a cabo por las penicilin-binding proteins (pbps), enzimas responsables de la elongaci\u00f3n y maduraci\u00f3n del peptidoglicano en el periplasma a trav\u00e9s de actividades, dd-carboxipeptidasa, dd-endoppetidassa, transpeptidasa, transglicosilada y beta-lactamasa. a trav\u00e9s del an\u00e1lisis in silico, nuestro grupo de trabajo descubri\u00f3 una pulativa nueva pbp, la cual debido a su homolog\u00eda con pbp4, se llam\u00f3 pbp4b. por esta raz\u00f3n uno de los principales objetivos es lograr el clonaje, expresi\u00f3n y purificaci\u00f3n de pbp4b a fin de determinar su funci\u00f3n biol\u00f3gica. la generaci\u00f3n de precursores solubles para la biosintesis del peptidoglicano y el proceso de divisi\u00f3n celular bacteriano involucra un grupo de genes localizados en el cluster dcw y que se hallan bajo el control del promotor pmra, con los genes mara y mraw presentes al inicio de dicho cluster. marw esta altamente conservado pero su funci\u00f3n biol\u00f3gica es desconocida. El clonaje, expresi\u00f3n y purificaci\u00f3n de mraw se realiz\u00f3 para determinar su funci\u00f3n en estos procesos bacterianos. los genes codificantes para pbp4b, mraz y mraw fueron inactivados en s l1344 utilizando una simple t\u00e9cnica basado en el proceso de recombinaci\u00f3n hom\u00f3loga.
