Las vacunas basadas en el polisac\u00e1rido capsular, aunque han sido exitosas contra el resto de serogrupos, no han podido dar respuesta a la prevenci\u00f3n de la enfermedad causada por dicho serogurpo, por lo que en la actualidad las investigaciones se centran en el estudio de otras mol\u00e9culas como las prote\u00ednas de membrana externa. No obstante, las exigentes caracter\u00edsticas que debe cumplir un ant\u00edgeno ideal con una reducida variabilidad antig\u00e9nica, exposici\u00f3n y expresi\u00f3n en la superficie o generaci\u00f3n de anticuerpos funcionales son requerimientos dif\u00edciles de cubrir por los candidatos propuestos hasta el momento. En estudios previos hemos detectado, entre diferentes ant\u00edgenos comunes al g\u00e9nero neisseria, un ant\u00edgeno de 47 kda (p47) capaz de genera anticuerpos en humanos tras la invasi\u00f3n del meningococo. En esta tesis doctoral nos hemos planteado identificarlo y estudiar sus caracter\u00edsticas moleculares, gen\u00e9ticas y antig\u00e9nicas en gran n\u00famero de cepas, as\u00ed como realizar una valoraci\u00f3n preliminar de su posible utilizaci\u00f3n en una vacuna universal contra dicha enfermedad. el ant\u00edgeno p47 se correspondi\u00f3 con una lipoprote\u00edna ubicada en la membrana externa de la envuelta del pat\u00f3geno bacteriano y tambi\u00e9n presente en n.Lactamica, n.Sicca y n.Subflava (gen nma0281, seg\u00fan cepa z2491 de n.Meningitis). Aunque no utilizamos ninguna cepa de n.Gonorrhoeae en nuestros estudios dicho gen tambi\u00e9n est\u00e1 presente en una cepa secuencia de esta especie (fa1090). El uso de diferentes medios de cultivo, el contexto gen\u00f3mico donde se ubica el gen y la similitud de dominios con otras prote\u00ednas conocidas nos indic\u00f3 que se trata de una mol\u00e9cula implicada en el metabolismo de metales.
