Al estudiar el efecto citot\u00f3xico de la fludarabina, la mitoxantrona y la ciclofosfamida sobre c\u00e9lulas primarias de lcm y sobre c\u00e9lulas de l\u00edneas celulares probadoras de la t(11;14) (q13;q32) se demuestra que la mitoxantrona, un inhibidor de la topoisomerasa ii, ejerce el mayor efecto citot\u00f3xico. Adem\u00e1s, la variante bl\u00e1stica es m\u00e1s sensible a la mitoxantrona que la variante t\u00edpica posiblemente debido a los elevados niveles de expresi\u00f3n de la topoisomerasa-ii. En los casos bl\u00e1sticos.  la citotoxicidad inducida por estos f\u00e1rmacos genot\u00f3xicos est\u00e1 mediada por la activaci\u00f3n de la v\u00eda apot\u00f3tica mitocondrial debido a un a\u00f1o en el dna, por lo que es necesario que los sensores de da\u00f1o a dna, p53 y\/o atm, sean funcionales.     el efecto dela fludarabina sobre c\u00e9lulas de llc-b viene determinado, en primera instancia, por el transporte v\u00eda hent2, atribuy\u00e9ndole importancia, por primera vez, en el efecto terap\u00e9utico de un an\u00e1logo de nucle\u00f3sidos.  la expresi\u00f3n de este transportador correlaciona significativamente con el efecto citot\u00f3xico inducido por la fludarabina en c\u00e9lulas de llc-b. Sin embargo, las c\u00e9lulas de lcm, que no responden in vitro a la fludarabina, expresan niveles m\u00e1s altos de mrna y de prote\u00edna de hent1 que de hent2. esta expresi\u00f3n diferencial podr\u00edan explicar la diferente respuesta a la fludarabina entre c\u00e9lulas de lcm y llc-b.
