{"id":53675,"date":"2018-03-09T22:41:23","date_gmt":"2018-03-09T22:41:23","guid":{"rendered":"https:\/\/www.deberes.net\/tesis\/sin-categoria\/caracterizacion-estructural-y-funcional-del-gen-kir2dl5\/"},"modified":"2018-03-09T22:41:23","modified_gmt":"2018-03-09T22:41:23","slug":"caracterizacion-estructural-y-funcional-del-gen-kir2dl5","status":"publish","type":"post","link":"https:\/\/www.deberes.net\/tesis\/quimica\/caracterizacion-estructural-y-funcional-del-gen-kir2dl5\/","title":{"rendered":"Caracterizaci\u00f3n estructural y funcional del gen kir2dl5."},"content":{"rendered":"<h2>Tesis doctoral de <strong> Natalia G\u00f3mez Lozano <\/strong><\/h2>\n<p>Los receptores kir (killer-cell immunoglobulin-like receptors), tras el reconocimiento de sus ligandos, las mol\u00e9culas de mhc clase i, regulan la respuesta de las c\u00e9lulas nk dirigida frente a c\u00e9lulas diana potenciales. los genes kir se localizan en la regi\u00f3n cromos\u00f3mica 19q13.4, en un complejo compuesto por hasta 15 genes y 2 pseudogenes, todos ellos con un alto grado de polimorfismo y formas alternativas de procesamiento.  kir2dl5 es el miembro de la familia descrito m\u00e1s recientemente, cuyo gen est\u00e1 presente s\u00f3lo en algunos individuos. Nuestro grupo ha estudiado su variabilidad intr\u00ednseca y su implicaci\u00f3n en la diversidad haplot\u00edpica de los genes kir. Este estudio ha identificado a kir2dl5 como un \u00abpunto caliente\u00bb de la diversidad kir, en el cu\u00e1l se han producido diferentes recombinaciones. hemos demostrado la existencia de dos loci que codifican para kir2dl5 (kir2dl5a y kir2dl5b), que pueden aparecer por separado o conjuntamente en el mismo haplotipo. Tambi\u00e9n hemos descubierto tres nuevos alelos de kir2dl5 (kir2dl5a*005, kir2dl5b*006 y *007) y un sobrecruzamiento desigual entre kir2dl5 y kir3dp1, que es presumiblemente el origen de dos haplotipos inusuales: aquellos portadores de una duplicaci\u00f3n de los genes conservados kir3dp1-kir2dl4-kir3dl\/s1 o una deleci\u00f3n de los mismos genes. Estos haplotipos nos han permitido descartar una funci\u00f3n esencial para kir2dl4, un receptor para hla-g, en reproducci\u00f3n, e identificar un alelo quim\u00e9rico funcional del pseudog\u00e9n kir3dp1 que podr\u00eda codificar para un receptor soluble.  la expresi\u00f3n de kir2dl5 es variable: tiene alelos transcritos en linfocitos nk y t y otros que se comportan como pseudogenes no transcritos. Esta divergencia se correlaciona con polimorfismos de las regiones promotoras. Hemos analizado la uni\u00f3n de factores de transcripci\u00f3n runx al promotor de los  alelos de kir2dl5, la actividad de estas regiones in vitro, y el efecto de una droga desmetilate en la
