Desarrollo de un programa de selecci\u00f3n cl\u00e1sica basado en mutaci\u00f3n al azar.  2. Clonaci\u00f3n de genes de b. Trispora implicados en la bios\u00edntesis de  -caroteno, y\/o que presenten una expresi\u00f3n constitutiva.  3. Desarrollo de un protocolo de transformaci\u00f3n de b. Trispora.  4. Expresi\u00f3n de genes biosint\u00e9ticos de  -caroteno en m. Circinelloides y b. Trispora. 5. Efecto de la luz sobre la producci\u00f3n de carotenoides en m. Circinelloides y b. Trispora.  se han identificado, clonado y caracterizado los genes acta, carb y carra. El orf del gen acta (1.561 pb) posee 4 intrones, codifica una g-actina, y su regi\u00f3n promotora es funcional en e. Coli y a. Chrysogenum. Los genes carb y carra se encuentran ligados en una regi\u00f3n de 4,5 kb del genoma de b. Trispora, y se transcriben en sentidos opuestos bajo el control de un promotor bidireccional de 611 pb. El orf del gen carb (1.955 pb) posee 2 intrones y codifica una prote\u00edna con actividad fitoeno deshidrogenasa. El orf del gen carra (1.894 pb) posee 1 intr\u00f3n y codifica una prote\u00edna con actividad fitoeno sintasa\/licopeno ciclasa. el gen carb de la cepa superproductora de ?-Caroteno b. Trispora f-744 posee la mutaci\u00f3n a1791c, que origina la sustituci\u00f3n del amino\u00e1cido ser presente en b. Trispora nrrl 2457 en la posici\u00f3n 528 por arg presente en f-744 (s528r). El gen carra de la cepa f-744 posee la mutaci\u00f3n t497c que origina la sustituci\u00f3n del amino\u00e1cido pro presente en nrrl 2457 en la posici\u00f3n 143 por ser presente en f-744 (p143s)). los genes carb y carra de b. Trispora son funcionales en m. Circinelloides. El gen carb de b. Trispora complementa la deficiencia en la actividad fitoeno deshidrogenasa de la cepa m. Circinelloides ms23, y el gen carra complementa la deficiencia en las actividades fitoeno sintasa y licopeno ciclasa de la cepa m. Circinelloides ms8. La expresi\u00f3n del gen carra de b. Trispora en m.
