Trwban70 constituye el dominio soluble de la prote\u00edna trwb, una prote\u00edna de membrana esencial en la conjugaci\u00f3n que acopla el relaxosoma (complejo de nucleoprote\u00edna) al sistema de transporte del dna en la conjunci\u00f3n del pl\u00e1smido r388. La estructura cristalogr\u00e1fica de trwban70 revela un hex\u00e1mero formado por seis subunidades id\u00e9nticas y un canal central. En este trabajo se caracteriza la actividad atpasa dependiente de dna de la prote\u00edna trwban70. La prote\u00edna presenta cooperatividad positiva para la hidr\u00f3lisis de atp con al menos tres sitios catal\u00edticos implicados en la hidr\u00f3lisis. La actividad es sensible al ph y la concentraci\u00f3n de sal, siendo m\u00e1s activa a valores bajos de ph. la longitud efectiva de dna necesaria para la activaci\u00f3n de la actividad a tpasa se encuentra entre 40 y 45 bases y \u00e9sta misma longitud es necesaria para la formaci\u00f3n de complejos trwban70-dna de elevado peso molecular, tal y como se observa mediante cromatograf\u00eda de gel filtraci\u00f3n. La mutaci\u00f3n de un residuo de tript\u00f3fano (w216a) situado en el canal central de la prote\u00edna origina una prote\u00edna defectiva en la hidr\u00f3lisis de atp. Por otro lado, este mutante ejerce un efecto dominante negativo tanto en la frecuencia de conjugaci\u00f3n del pl\u00e1smido r388 como en la hidr\u00f3lidis de atp, avalando el estado oligom\u00e9rico de la prote\u00edna. La hidr\u00f3lisis de atp no se encuentra acoplada a una actividad helicasa en las condicones ensayadas. Por otro lado trwban70 interact\u00faa con el relaxosoma mediante la prote\u00edna trwa, que se ha observado estimula la hidr\u00f3lisis de atp de trwban70 mediante contactos prote\u00edna-prote\u00edna. Estos resultados, avalados adem\u00e1s por la similitud estructural de trwban70 con el complejo f1-atpasa, nos llevan a proponer una funci\u00f3n<\/p>\n<p>&nbsp;<\/p>\n<h3>Datos acad\u00e9micos de la
