En este trabajo hemos tratado de contrarrestar la disfunci\u00f3n mitocondrial  mediante estrategias de transferencia g\u00e9nica con vectores de hsv-1, con el objetivo de paliar la neurodegeneraci\u00f3n.     en primer lugar, hemos establecido modelos experimentales de c\u00e9lulas neuronales para llevar a cabo estudios de neurodegeneraci\u00f3n y neuroprotecci\u00f3n. S\u00ed, hemos empleado el agente neurot\u00f3xico rotenona para inducir disfunci\u00f3n mitocondrial a trav\u00e9s de la inhibici\u00f3n del complejo i mitocondrial, constituyendo un modelo experimental interesante para evaluar la influencia de la disfunci\u00f3n mitocondrial en la neurodegeneraci\u00f3n.     en segundo lugar hemos estudiado el efecto de modular la actividad de la enzima gluc\u00f3geno sintasa quinasa-3 (gsk-3) sobre la toxicidad inducida por rotenona, ya que la gsk-3 es una quinasa implicada en la modulaci\u00f3n de distintos procesos celulares, incluyendo la apoptosis. Demostramos que la inhibici\u00f3n cr\u00f3nica de la gsk-3, bien mediante la expresi\u00f3n de un mutante dominante negativo de gsk-3 (85r), bien mediante el empleo de inhibidores farmacol\u00f3gicos, tiene un efecto neuroprotector frente a la rotenona en c\u00e9lulas similares a neuronas humanas en cultivo y en neuronas primarias de bulbo raqu\u00eddeo de rat\u00f3n. Hemos evaluado distintos mecanismos moleculares posibles que expliquen este efecto neuroprotector. Mostramos que la inhibici\u00f3n cr\u00f3nica de gsk-3 resulta en un incremento en el consumo de glucosa por la v\u00eda glucol\u00edtica. Adem\u00e1s, hemos observado tambi\u00e9n que la neuroprotecci\u00f3n inducida por inhibici\u00f3n cr\u00f3nica de gask-3, la hexoquinasa ii (hkii) se localiza preferentemente en la mitocondrial mientras que el transportador de glucosa glut3 se transloca a la membrana plasm\u00e1tica.
