Por lo tanto, se consider\u00f3 estudiar la funci\u00f3n de la actina en este tejido mediante la ptd4-pfnil como herramienta molecular para interferir con la din\u00e1mica del citoesqueleto de actina. La secu\u00e9ncia ptd4 (ho et al., 2001) es un p\u00e9ptido de transducci\u00f3n, permeable a membrana que, fusionado con pfn, le permite acceder al interior celular. Profilina (pfn) es una prote\u00edna que une mon\u00f3meros de actina, le intercambia el adp con atp y presenta la actina-atp al extremo + de un filamento de actina durante la polimerizaci\u00f3n. Pfn se encuentra en dominios intracelulares donde la actina es muy din\u00e1mica, como filopodios y lamelipodios.     primero, se crearon vectores de expansi\u00f3n para la producci\u00f3n de prote\u00ednas de fusi\u00f3n con ptd4, se clonaron en ellos los genes de profilina i (pfnl: ubicua) y profilina ii (pfnii; neuronal), as\u00ed como de un marcador como *-galactosidasa, y se purificaron las prote\u00ednas de fusi\u00f3n resultantes. despu\u00e9s se comprob\u00f3 la transducci\u00f3n de estas prote\u00ednas en c\u00e9lulas trabeculares y su dependencia  del tiempo y de la concentraci\u00f3n, as\u00ed como su independencia de la temperatura. en c\u00e9lulas trabeculares en cultivo, se observ\u00f3 que ptd4-pfnl induc\u00eda la formaci\u00f3n de lamelipodios fue similar a la producida pro suero o factores de crecimiento, tanto por la presencia de marcadores de lamelipodios (arp2\/3 y rac) como por la ausencia de lamelipodios tras la inhibici\u00f3n de diferentes componentes de la v\u00eda de se\u00f1alizaci\u00f3n (rac y pi4k) o de la contractibilidad (miosina y mlck).  este efecto de ptd4-pfnil depend\u00eda tanto de la uni\u00f3n
