{"id":55200,"date":"2018-03-09T22:42:55","date_gmt":"2018-03-09T22:42:55","guid":{"rendered":"https:\/\/www.deberes.net\/tesis\/sin-categoria\/efectos-anti-inflamatorios-de-derivados-sinteticos-del-acido-acantoico\/"},"modified":"2018-03-09T22:42:55","modified_gmt":"2018-03-09T22:42:55","slug":"efectos-anti-inflamatorios-de-derivados-sinteticos-del-acido-acantoico","status":"publish","type":"post","link":"https:\/\/www.deberes.net\/tesis\/bioquimica\/efectos-anti-inflamatorios-de-derivados-sinteticos-del-acido-acantoico\/","title":{"rendered":"Efectos anti-inflamatorios de derivados sint\u00e9ticos del \u00e1cido acantoico"},"content":{"rendered":"<h2>Tesis doctoral de <strong> Francisca Gonz\u00e1lez Trav\u00e9s <\/strong><\/h2>\n<p>En los terpenos constituyen una amplia familia de esteroides naturales ampliamente distribuidos en plantas e insectos.  en el presente estudio hemos analizado el efecto de derivados sint\u00e9ticos del \u00e1cido acantoico en la respuesta de macr\u00f3fagos ante est\u00edmulos proinflamatorios.  estos diterpenos pimaranos inhiben selectivamente la activaci\u00f3n dela v\u00eda de se\u00f1alizaci\u00f3n ikk\/nfkb y no parecen afectar a la activaci\u00f3n de otras map quinasas como erk y p38. El an\u00e1lisis de las posibles dianas de estos diterpenos se\u00f1ala a nik como la principal responsable de la inhibici\u00f3n de ikk observada, as\u00ed como de la reducci\u00f3n de fosforilaci\u00f3n y degradaci\u00f3n de ikbalfa. En la administraci\u00f3n in vivo de los diterpenos se observ\u00f3 una reducci\u00f3n del edema inducido por tpa en oreja de rat\u00f3n, una disminuci\u00f3n de los niveles de tnfalfa en plasma y un aumento de la supervivencia de los ratones en un modelo de da\u00f1o hep\u00e1tico inducido por lps\/d-galn. todos estos datos apuntan a nik como la principal diana de los diterpenos e indican que \u00e9stos tienen actividad anti-inflamatoria in vivo. Adem\u00e1s, hemos investigado los efectos de estas mol\u00e9culas sobre la activaci\u00f3n de los receptores nucleares comprobando que estos diterpenos son potentes activadores de los liver x receptors (lxralfa y lxrbeta) tanto en l\u00edneas celulares macrof\u00e1gicas de origen humano y murico como en cultivos primarios de macr\u00f3fagnos peritoneales y derivados de m\u00e9dula \u00f3sea. \u00e9stos compuestos aumentaron de forma significativa el flujo de colesterol desde el macr\u00f3fago. De hecho, la activaci\u00f3n de macr\u00f3fagos con los diterpenos indujo la expresi\u00f3n de los genes diana de lxr y el flujo de colesterol con una efectividad similar a la delos agonistas sint\u00e9ticos de los lxrs. Adem\u00e1s, los efectos observados depreciaron en macr\u00f3fagos procedentes de ratones carentes de ambas isoformas (lxralfa, beta-\/-). los resultados obtenidos muestran la capacidad de estos
