Sus acciones est\u00e1n mediadas por interacciones con receptores de membrana celular acoplados a prote\u00ednas g, de los cuales se han clonado cuatro subtipos: a1, a2a, a2b y a3. El receptor a2b es el menos estudiado, quiz\u00e1 por su baja afinidad por la adenosina y la falta de pruebas farmacol\u00f3gicas espec\u00edficas. En los \u00faltimos a\u00f1os ha aumentado considerablemente el inter\u00e9s por este subtipo de receptor al comprobarse que est\u00e1 implicado en importantes funciones, como la secreci\u00f3n g\u00e1strica de \u00e1cido. en esta tesis se han utilizado c\u00e9lulas parietales de mucosa g\u00e1strica de conejo y membranas aisladas para caracterizar el receptor a2b adenosina. Se ha observado que la ocupaci\u00f3n del receptor a2b de adenosina por sus agonistas neca y 2-cado en las c\u00e9lulas parietales aisladas de la mucosa g\u00e1strica de conejo estimula las secreci\u00f3n de \u00e1cido mediante la activaci\u00f3n de la adenilato ciclasa, pero no produce modificaciones de los niveles de ca2+ citos\u00f3lico. Los experimentos de microscop\u00eda confocal y citometr\u00eda de flujo indican que el receptor a2b de adenosina y la adenosina desaminasa colocalizan parcialmente en las membranas de estas c\u00e9lulas, especialmente en los lugares de contacto de unas c\u00e9lulas con otras. Mediante western blotting y extracci\u00f3n selectiva de prote\u00ednas de membrana se demuestra que la grp94 est\u00e1 tambi\u00e9n presente como prote\u00edna no integral en la membrana de la c\u00e9lula parietal. Los experimentos de uni\u00f3n a radiolingados indican que existen dos lugares de uni\u00f3n a [3h]-neca en las membranas de estas c\u00e9lulas: el receptor a2b de adenosina y la grp94. El radicicol inhibe la uni\u00f3n de [3h]-neca a las membranas son una ki 19 nm.
