{"id":5558,"date":"1995-01-01T00:00:00","date_gmt":"1995-01-01T00:00:00","guid":{"rendered":"https:\/\/www.deberes.net\/tesis\/1995\/01\/01\/apoptosis-en-celulas-hematopoyeticas-regulacion-de-la-actividad-endonucleasa\/"},"modified":"1995-01-01T00:00:00","modified_gmt":"1995-01-01T00:00:00","slug":"apoptosis-en-celulas-hematopoyeticas-regulacion-de-la-actividad-endonucleasa","status":"publish","type":"post","link":"https:\/\/www.deberes.net\/tesis\/quimica\/apoptosis-en-celulas-hematopoyeticas-regulacion-de-la-actividad-endonucleasa\/","title":{"rendered":"Apoptosis en celulas hematopoyeticas. regulacion de la actividad endonucleasa."},"content":{"rendered":"<h2>Tesis doctoral de <strong> Rosalia Ascaso Lopez De Soria <\/strong><\/h2>\n<p>En esta tesis doctoral se ha realizado el estudio de las caracteristicas de la muerte celular programada en una linea de celulas hematopoyeticas procedentes de celula osea de raton y que son dependientes de interleuquina 3 para sobrevivir y proliferar, desencadenandose la apoptosis tras retirada de la citoquina detectable a las 15h de cultivo en ausencia de este factor.La inhibicion de la topoisomerasa ii en estas celulas mediante el tratamiento con etoposido o vp16, compuesto utilizado en quimioterapia, produce una aceleracion de la apoptosis tras retirada de il3 en esta linea celular mediante un mecanismo dependiente de la sintesis de proteinas y de arns e inhibible mediante adicion del inhibidor general de nucleasas conocido como acido aurin tricarboxilico (ata). La adicion tardia de il3 es capaz de prevenir la apoptosis tras adicion del etoposido hasta tiempos muy cercanos en los que se detecta la fragmentacion en \u00abescalera\u00bb tipica de la apoptosis por lo que este debe ser uno de los puntos irreversibles en el mecanismo de accion del etoposido.  celulas ba\/f3 que sobreexpresan bcl-2 son mas resistentes a la apoptosis inducida por el etoposido mediante un mecanismo independiente pero a su vez cooperativo con el utilizado por la interleuquina 3.  la apotosis inducida por el etoposido es independiente de un aumento en la expresion de la proteina supresora de tumores o p53.  en extractos proteicos nucleares de celulas ba\/f3 se ha detectado una actividad endonucleasa dependiente de calcio y regulable por iones monovalentes. Tras purificacion de este extracto mediante cromatografia de alta afinidad se ha detectado una proteina de 30 kda con actividad endonucleasa regulable por iones monovalentes, dependiente de calcio a ph 7.0 e independiente a ph&lt;6.5.  la acidificacion intracelular en condiciones farmacologicas induce apoptosis de las celulas ba\/f3 en presencia de il3 y es inhibida tras cultivo en un medio con
