Aunque la tuberculosis es una enfermedad tratable con antibi\u00f3ticos y puede prevenirse mediante vacunaci\u00f3n, la aparici\u00f3n de cepas resistentes a los f\u00e1rmacos antituberculosos y la variable eficacia protectora de la vacuna bcg contra las manifestaciones pulmonares de la enfermedad dejan la victoria sobre la tuberculosis fuera de alcance.  la b\u00fasqueda de una vacuna eficaz contin\u00faa, iniciativas conjuntas de laboratorios europeos y americanos han hecho posible la construcci\u00f3n de eficaces candidatos a vacuna. De hecho, el punto de partida de \u00e9ste trabajo ha sido el gen phop, cuyo papel en la virulencia de m. Tuberculosis fue estudiado previamente en nuestro laboratorio en colaboraci\u00f3n con el instituto pasteur de par\u00eds. Un mutante phop construido en la cepa cl\u00ednica mt103 confiere mejor protecci\u00f3n contra la tuberculosis que la actual vacuna bcg en varios modelos animales, estos resultados apuntan a dicho mutante como un prometedor candidato a vacuna viva atenuada. Sin embargo, aunque el fenotipo virulento del mutante phop ha sido bien estudiado, los mecanismos moleculares que conducen a la atenuaci\u00f3n no est\u00e1n caracterizados. Por ello, esta tesis se ha enfocado a comprender la funci\u00f3n del gen phop y descifrar su papel en la virulencia del bacilo de la tuberculosis.  phop es un regulador transcripcional que forma parte del sistema de dos componentes (2cs) phopr de m. Tuberculosis. Los 2css median cambios transcripcionales en respuesta a determinados est\u00edmulos y est\u00e1n implicados en la regulaci\u00f3n de la virulencia en bacterias pat\u00f3genas. Con el objetivo de caracterizar gen\u00e9ticamente el papel del sistema phopr, hemos construido mutantes por delecci\u00f3n en el gen phop o en ambos genes phopr en tres cepas diferentes de
