Las quinasas citoplasm\u00e1ticas mejor estudiadas son la familia de tirosina quinasas src (sfk) y jak2. Previamente, se ha observado en el laboratorio que la activaci\u00f3n de c-src por prl, no requiere la activaci\u00f3n de jak2 y que, adem\u00e1s de la actividad enzim\u00e1tica de la sfk, la capacidad adaptadora de esta familia est\u00e1 implicada en la se\u00f1alizaci\u00f3n celular inducida por prl.     en este trabajo, en primer lugar, hemos estudiado si la activaci\u00f3n de jak2, considerada el mayor efector en la se\u00f1alizaci\u00f3n por prl, requiere la capacidad adaptadora dela sfk, nuestros resultados muestran que la sfk, a trav\u00e9s de sus dominio adaptadores regula la activaci\u00f3n de jak2, mientras que su actividad quinasa controla la estimulaci\u00f3n de la v\u00eda pl3l-akt y p70s6k y la expresi\u00f3n de c-myc. La quinasa jak2 regula la activaci\u00f3n de las v\u00edas jak2\/stat5 y mek1\/2-erk1\/2, pero no est\u00e1 implicada en el control de la estimulaci\u00f3n del a sfk, demostrando que la sfk es el mayor efector en la se\u00f1alizaci\u00f3n por prl. Por otro lado, la tirosina fosfatasa shp2 se comporta como enlace entre la sfk y jak2, ya que regula la se\u00f1alizaci\u00f3n mediada por jak2 pero sin alterar la activaci\u00f3n de la sfk. La interacci\u00f3n entre shp2 y la sfk regula la activaci\u00f3n de jak2 por prl, demostrando la importancia de la capacidad adaptadora de la sfk en la se\u00f1alizaci\u00f3n por prl.     en segundo lugar, hemos analizado los mecanismos moleculares por lo que la v\u00eda sfk\/pl3\/akt regula la expresi\u00f3n de c-myc. Nuestros resultados demuestran que la actividad de akt regula los niveles dec-myc, controlando su expresi\u00f3n a nivel transcripcional por medio de la inactivaci\u00f3n del factor de  transcripci\u00f3n \u00abforkhead\u00bb fkhrl1 y su estabilidad, bloqueando su degradaci\u00f3n mediada por la gluc\u00f3geno sintetasa qui<\/p>\n<p>&nbsp;<\/p>\n<h3>Datos acad\u00e9micos de la tesis
