Estos mecanismos se conocen como checkpoints y pueden ser activados por da\u00f1os en el dna, problemas en la replicaci\u00f3n del dna o en la alineaci\u00f3n de los cromosomas metaf\u00e1sicos. En esta tesis se ha estudiado el checkpoint que se activa cuando se inhibe la replicaci\u00f3n con hidroxiurea (hu) en c\u00e9lulas de mam\u00edfero, comparando c\u00e9lulas tumorales y no tumorales. En el laboratorio y se hab\u00eda descrito la capacidad de las c\u00e9lulas no tumorales nrk y niht3t3 de activar el checkpoint de replicaci\u00f3n a pesar de la inhibici\u00f3n con cafe\u00edna de las principales quinasas de respuesta descritas hasta el momento, atr y atm. Sin embargo hab\u00eda disparidad de respuestas entre las c\u00e9lulas tumorales habiendo algunas capaces de evitar la entrada en mitosis en presencia de hu (respuesta s\/m) al inhibir atr y atm (hela, sw480) y otras en cambio incapaces de llevar a cabo esta parada (hct116, ht29, caco2). En esta tesis se demuestra que las c\u00e9lulas tumorales hela responden a hu independientemente de atr y atm pero dependientemente de la activaci\u00f3n de chk1 de manera distinta a la cl\u00e1sicamente descrita fosforilaci\u00f3n de la serina 345 y 317. Adem\u00e1s dicha activaci\u00f3n del checkpoint no depende de dna-pk ni de ninguna pl3k quedando pro determinar qu\u00e9 tipo de modificaci\u00f3n posttraduccional regular\u00eda a chk1 y por parte de qu\u00e9 prote\u00ednas. En estas c\u00e9lulas hemos analizado la importancia de prote\u00ednas sensores y mediadoras de la v\u00eda y hemos visto que la presencia de rad17  no es esencial para la respuesta dependiente e independiente de atr\/atm pero sin embargo la mediadora claspina es importante para la respeta s\/m del checkpoint de replicaci\u00f3n cuando se inhibe atr\/atm, vi\u00e9ndose una relaic\u00f3n entre esta prote\u00edna y la regulaci\u00f3n a la baja  de la ciclina b1, importante para la entrada en mitosis.
