Gema Elvira <\/strong><\/h2>\n<p>La prote\u00edna del prion celular, prpc se ha convertido en una molecular clave en las enfermedades de priones dado su papel precursor de conf\u00f3rmeros auto-perpetuantes. Con el fin de delinear la ruta de conversi\u00f3n de prpc en prpsc, hemos encontrado que una leve perturbaci\u00f3n estructural en medio \u00e1cido, copatible con las condicones fisiol\u00f3gicas, induce la transformaci\u00f3n de alfa-rshaprp(2-232) en un estado compacto, rshaprp*(23-232), competente para la agregaci\u00f3n. esta competencia para la agegaci\u00f3n se manifiesta en estados aglomerados, simulados por la inclusi\u00f3n de pol\u00edmeros inertes de peg, como una polimerizaci\u00f3n nucleada conformacionalmente que implica a varias formas ricas en l\u00e1mina beta. Los pol\u00edmeros comparten con prpsc33-35 la insolubilidad, la carencia de orden macrosc\u00f3pico y su capacidad para generar n\u00facleos de resistencia a proteolisis cuando se unen a cu(ii). Sin embargo carecen de infectividad in vivo, sugiriendo que muchas propiedades atribuidas diferencialmente a prpsc corresponden a caracter\u00edsticas fisiol\u00f3gicas de prpc. Adem\u00e1s, de esta multiplicidad conformacional, en prp se ha descrito una variabilidad topol\u00f3gica, producto de la cual se generan cuatro formas b\u00e1sicas, secprp, ntmprp, ctmprp y cyprp. Dentro de ellas, cyprp representa la poblaci\u00f3n de cadenas nacientes que se segregan fuera de la ruta de secreci\u00f3n, en un proceso dependiente de la secuencia del p\u00e9ptido se\u00f1al.  en este trabajo mostramos que cyprp corresponde a una isoforma, prp(1-14), resultante de la iniciaci\u00f3n de la traducci\u00f3n en un sitio secundario.  esta forma presenta una distribuci\u00f3n nucleocitoplasm\u00e1tica y se detecta en tejidos cerebrales normales. su expresi\u00f3n desacoplada del resto de la familia de prp, interfiere con la proliferaci\u00f3n celular.  estos resultados apuntan a que la funci\u00f3n de prpc es el resultado de la coordinaci\u00f3n de las funciones de cada forma y, que la patolog\u00eda podr\u00eda ser el resultado de la p\u00e9rdida de dicha coordinaci\u00f3n.<\/p>\n<p>&nbsp;<\/p>\n<h3>Datos acad\u00e9micos de la tesis doctoral \u00ab<strong>Diversidad molecular y plasticidad conformacional de la prote\u00edna del prion celular<\/strong>\u00ab<\/h3>\n<ul>\n<li><strong>T\u00edtulo de la tesis:<\/strong>\u00a0 Diversidad molecular y plasticidad conformacional de la prote\u00edna del prion celular <\/li>\n<li><strong>Autor:<\/strong>\u00a0  Serrano M.
