Dicha regulaci\u00f3n se encuentra condicionada por la estructura de la cromatina, que a pesar del hecho de ser din\u00e1mica, supone un obst\u00e1culo para la uni\u00f3n de los direrentes componentes de la maquinaria transcripcional al dna. Para resolver estas restricciones, la c\u00e9lula ha organizado el reclutamiento de enzimas modificadores de la cromatina y de complejos remodeladores de la cromatina dependientes de atp. El primer grupo de enzimas son las encargadas de modificar covalentemente las historias, alterando generalmente la carga de las mismas y produciendo el reclutamiento de cofactores espec\u00edficos. La actividad del segundo grupo de enzimas consiste en utilizar la energ\u00eda procedente de la hidr\u00f3lisis del atp para modificar las interacciones historia-adn, incrementando con ello la accesibilidad del adn nucleosomial a la maquinaria transcripcional. La comprensi\u00f3n de los mecanismos que utiliza la c\u00e9lula para regular la expresi\u00f3n de un gen inmediato-temprano, como egr-1, es de gran importancia tanto para el conocimiento b\u00e1sico molecular como para la comprensi\u00f3n de diferentes patolog\u00edas. La acci\u00f3n coordinada de las diferentes cascadas de se\u00f1alizaci\u00f3n, la uni\u00f3n de factores transcripcionales y de complejos enzim\u00e1ticos que act\u00faan sobre la cromatina es fundamental para la correcta interpretaci\u00f3n del c\u00f3digo gen\u00e9tico, que depende de cada gen concreto. En el caso del gen egr-1, el tratamiento de c\u00e9lulas mlp29 con tpa activa la expresi\u00f3n a los 5 min., Alcanza un pico entre los 15 y 30min del tratamiento y deja de ser detectable a los 180min. Dicha inducci\u00f3n, se produce a trav\u00e9s de las cascadas de quinasas mek y p38, que adem\u00e1s act\u00faan fosfofilando los factores elk1 y creb, presentes en el promotor del gen.
