Sin embargo, a concentraciones peque\u00f1as  act\u00faan como mensajeros intracelulares, capaces de desencadenar cascadas de se\u00f1alizaci\u00f3n que conducen a la activaci\u00f3n de enzimas y de factores de transcripci\u00f3n, alterando finalmente la expresi\u00f3n gen\u00e9tica y mediando respuestas celulares de adaptaci\u00f3n o de muerte celular. La presente tesis doctoral propone elucidar los mecanismos de se\u00f1alizaci\u00f3n dirigidos por el agente antitumoral doxorrubicina (dox) en cultivos primarios de hepatocitos, las respuestas que provoca a largo plazo, as\u00ed como establecer la implicaci\u00f3n de especies radicalarias en dichas respuestas. Como resultado de este trabajo se concluye que la dox induce se\u00f1alizaci\u00f3n intracelular sin provocar estr\u00e9s oxidativo ni citotoxidaid, activando las v\u00edas de las mapks erl, p38 y jnk depende de la generaci\u00f3n de ros, en particular de ani\u00f3n super\u00f3xido, siendo p38 activada por mecanismos independientes de ros. La dox provoca la movilizaci\u00f3n de nf-kb que depende de la generaci\u00f3n de ani\u00f3n super\u00f3xido y que cursa de una forma at\u00edpica (tanto pro su cin\u00e9tica como por su persistencia), no vi\u00e9ndose acompa\u00f1ada de aumento de la actividad transcripcional, sino m\u00e1s bien de un comportamiento represor sobre los genes con secuencias -kb en sus promotores. En la v\u00eda de movilizaci\u00f3n de nf-kb no participa ninguna de las mapks analizadas, pero s\u00ed alguna de las isoformas de pkc, siendo su activaci\u00f3n dependiente de ani\u00f3n super\u00f3xido.
