Las glicoprote\u00ednas virales (gp120 y gp41) de la envuelta (env) de vih se sintetizan en el ret\u00edculo endopl\u00e1smico (re) como un precursor de 160 kilodaltons que se procesa proteol\u00edticamente en el aparato de golgi por proteasas celulares. En nuestro laboratorio se ha desarrollado una estrategia para interferir con la funci\u00f3n de env. Una quimera de cd4 (cd4e15) que contiene una se\u00f1al de retenci\u00f3n en el re derivada del dominio citopl\u00e1smico de cd3e ha sido utilizada para interferir con la salida de gp160 del re. Cd4e15 queda retenida en ese compartimento e impide el procesamiento y maduraci\u00f3n de gp160 a gp120 y gp41.  el modo de acci\u00f3n de cd4e15 es m\u00e1s complejo dado que la expresi\u00f3n de la quimera inhibe la replicaci\u00f3n de vih incluso cuando la quimera se expresa en una minoria de c\u00e9lulas. Esto sugiri\u00f3 la existencia de un factor antiviral producido por las c\u00e9lulas transducidas, protegiendo a las c\u00e9lulas no transducidas, efecto que se ha podido corroborar por experimentos en placas de doble pocillo separados por un poro de 0,4 micras. La liberaci\u00f3n del factor depende de la infecci\u00f3n. En este trabajo estudiamos al detalle que puede ser ese factor soluble antiviral.  Finalmente los resultados indican que el factor soluble antiviral es un fragmento de la prote\u00edna viral gp120. La  identificaci\u00f3n del factor ha sido posible tanto mediante t\u00e9cnicas bioqu\u00edmicas as\u00ed como t\u00e9cnicas de prote\u00f3mica. La identificaci\u00f3n de un nuevo factor antiviral abre la puerta a nuevas terapias para el tratamiento del vih.<\/p>\n<p>&nbsp;<\/p>\n<h3>Datos acad\u00e9micos de la tesis doctoral
