Pombe posee una morfolog\u00eda cil\u00edndrica cuyo mantenimiento depende en gran medida de la presencia de una pared celular externa a su membrana plasm\u00e1tica (brunner and nurse, 2000). La pared celular de los hongos y levaduras esta formada por glicoprote\u00ednas y polisac\u00e1ridos (\u00c2\u00bf y \u00edY-glucanos y quitina). En s. Pombe el componente mayoritario y el primer polisac\u00e1rido que se deposita es el \u00edY-(1,3)-glucano (45-48% de la pared) (duran and perez, 2004).  en nuestro laboratorio se han aislado una serie de mutantes sensibles a equinocandina, un compuesto antif\u00fangico que inhibe espec\u00edficamente la actividad \u00edY-(l ,3)-gluc\u00e1n sintasa (carnero et al., 2000). Uno de estos mutantes, denominado ehs2-1, adem\u00e1s es termosensible, a 36\u00c2\u00b0c las c\u00e9lulas se lisan, principalmente durante el proceso de divisi\u00f3n celular. Ehs2-1 presenta una actividad \u00edY-(1,3)-gluc\u00e1n sintasa reducida, hasta casi el 50% a la temperatura restrictiva comparado con una cepa silvestre. Mediante mapeo gen\u00e9tico, encontramos que la mutaci\u00f3n de ehs2-1 se hallaba asociada al marcador ade6+ en el cromosoma iii (garc\u00eda, blanca. Tesis doctoral, 2002). Analizando los genes cercanos a esta regi\u00f3n se encontraron dos orfs, contiguas en el cromosoma, que podr\u00edan tener relaci\u00f3n con la bios\u00edntesis de la pared celular. Estas orfs, denominadas rgf1+ y rgf3+, poseen un dominio rhogef presente en todos los reguladores positivos de las gtpasas de la familia rho. En s. Pombe hay seis prote\u00ednas que pertenecen a la familia rho, rholp, rho2p, rho3p, rho4p, rho5p y cdc42p, de ellas solamente rholp (la subunidad reguladora de la actividad \u00edY-(1,3)-gluc\u00e1n sintasa) y rho2p se han relacionado con la bios\u00edntesis de la pared celular (arellano et al., 1997; arellano et al., 1996; calonge et al., 2000).  hemos visto que rgf3+, donado en un pl\u00e1smido multicopia, rescata todos los fenotipos del mutante ehs2-1, lisis a 36\u00c2\u00b0c e hipersensibilidad a equinocandina, mientras que en las mismas condiciones rgf1+ no complementa apenas el fenotipo de lisis, y solo es capaz de rescatar parcialmente el fenotipo de hipersensibilidad a antif\u00fangicos, lo que parece indicar que es rgf3+ el gen afectado en el mutante ehs2-1.  en estudios anteriores se hab\u00eda visto que las c\u00e9lulas que presentan niveles muy bajos de rholp tambi\u00e9n se lisan (arellano et al., 1996).
