La disecci\u00f3n de sus mecanismos de acci\u00f3n permitir\u00e1 la creaci\u00f3n de estrategias que permitan potenciar los efectos beneficiosos frente a los adversos. Los glucorticoides ejercen parte de sus acciones a trav\u00e9s de la inhibici\u00f3n de la v\u00eda de las mapks, v\u00edas activadas por est\u00edmulos proinflamatorios, entre otros. La mkp-1 es una fosfatasa dual capaz de fosforilar, inactivando, a las mapks y cuya expresi\u00f3n aumenta por dexametasona. Constituye, pues, un mecanismo de interacci\u00f3n entre las v\u00edas de los glucorticoides y de las mapks.     en este trabajo se ha estudiado la regi\u00f3n reguladora del gen de la mkp-1 en busca de elementos de respuesta a glucocorticoides (gre) funcionales que demostrasen que la inducci\u00f3n por glucocorticoides de este gen es mediada pro su receptor (gr), identific\u00e1ndose tres gres funcionales en la regi\u00f3n reguladora proximal (-2000 pb) del gen de rat\u00f3n, responsables de la respuesta de esta regi\u00f3n a glucocorticoides.     una vez comprobada la inducibilidad directa por glucocorticoides de este gen, se estudi\u00f3 su participaci\u00f3n en las acciones antiinflamatorias de los glucocorticoides en diferentes modelos experimentales sobre ratones deficientes en esta fosfatasa. El papel de la mkp-1 depend\u00eda del modelo usado. en inflamaci\u00f3n local, los ratones deficientes en mkp-1 perd\u00edan sensibilidad ante los glucocorticoides: dosis capaces de revertir la inflamaci\u00f3n en salvajes, ten\u00edan un efecto menor en los ratones deficientes, y un efecto intermedio en los heterocigotos. En inflamaci\u00f3n sist\u00e9mica, por endotoxemia, sin embargo, los glucocorticoides revert\u00edan completamente la mortalidad asocaida a lps, de manera totalmente independiente de mkp-1. Tambi\u00e9n se observan diferencias en cuanto a los tipos celulares analizados y al est\u00edmulo proinflamatorio usado.
