Varios estudios sugieren un importante papel de la endotelina-1 (et-1), la angiotensina ii (ang ii) y el factor transformador del crecimiento (tgf- ) en enfermedades vasculares. El primer objetivo de esta tesis fue estudiar si la et-1 regula ctgf. En cmlv obtenidas de aorta de rata la et-1 aument\u00f3 de manera significativa la expresi\u00f3n de arnm de ctgf, la actividad del promotor de ctgf y la producci\u00f3n de prote\u00edna a trav\u00e9s del receptor eta. El bloqueo de ctgf, con un oligonucle\u00f3tido antisentido, disminuy\u00f3 la producci\u00f3n de fibronectina y col\u00e1geno tipo i inducidos por et-1. El segundo objetivo fue determinar si existe interacci\u00f3n entre et-1, ang ii y tgf-\u00edY en procesos de fibrosis vascular y sus mecanismos intracelulares. En cmlv la et-1 a trav\u00e9s del receptor y de la ruta de rhoa\/quinasa de rho induce la producci\u00f3n de ctgf. La regulaci\u00f3n de ctgf por et-1 es independiente de tgf-\u00edY, ya que la et-1 no indujo la expresi\u00f3n g\u00e9nica ni la producci\u00f3n de tgf-\u00edY y adem\u00e1s la producci\u00f3n de ctgf por et-1 es indepediente de tgf- , mostrando diferencias con ang ii. En esta tesis encontramos una relaci\u00f3n directa entre et-1 y ang ii, ya que el antagonista del receptor eta disminuy\u00f3 la inducci\u00f3n de ctgf dependiente de ang ii, esto podr\u00eda indicar que la et-1 es mediadora de las acciones de ang ii. El tgf-\u00edY regula procesos de fibrosis actuando a trav\u00e9s de la ruta de las prote\u00ednas smad. Nuestros resultados demuestran que la ang ii activa las prote\u00ednas smad en c\u00e9lulas vasculares. Las prote\u00ednas smad est\u00e1n implicadas en la sobreexpresi\u00f3n de ctgf y de prote\u00ednas de mec inducidas por la angii, independientemente del tgf-\u00edY. Este nuevo hallazgo sugiere que la activaci\u00f3n de smad podr\u00eda estar implicada en los efectos profibrog\u00e9nicos de la angii en enfermedades vasculares.<\/p>\n<p>&nbsp;<\/p>\n<h3>Datos
