El \u00fanico m\u00e9todo de diagn\u00f3stico de vhvs reconocido oficialmente por la uni\u00f3n europea est\u00e1 basado en la detecci\u00f3n del agente viral en cultivo celular seguido de la identificaci\u00f3n mediante t\u00e9cnicas inmunol\u00f3gicas. En esta tesis se propone la rt-pcr como m\u00e9todo de detecci\u00f3n e identificaci\u00f3n alternativo o complementario al cultivo celular. Para ello, se ha llevado a cabo la optimizaci\u00f3n de la t\u00e9cnica en todos sus pasos, incluyendo la extracci\u00f3n del \u00e1cido nucleico y la selecci\u00f3n de cebadores. Los resultados obtenidos permitieron concluir que se ha conseguido poner a punto una metodolog\u00eda r\u00e1pida, sensible (l\u00edmite de detecci\u00f3n 10 fg de arn viral) y altamente espec\u00edfica para el diagn\u00f3stico de este virus. La sensibilidad del ensayo in vitro se ha testado usando rna procedente de tejidos infectados experimentalmente. El l\u00edmite de detecci\u00f3n de la rt-pcr fue estimado en 1\u00ed&#8211;10exp0 o 0,97\u00ed&#8211;10exp3 tcid50\/g (dependiendo de si la adsorci\u00f3n viral ha tenido lugar durante 1 o 24 horas, respectivamente). La confirmaci\u00f3n de los resultados de rt-pcr, tanto mediante southern blot como por nested-pcr, incrementaron la sensibilidad de la t\u00e9cnica de detecci\u00f3n en 1 o 2 logaritmos, respectivamente. tambi\u00e9n, se ha desarrollado un m\u00e9todo incruento para la detecci\u00f3n del virus vhsv basado en la tecnolog\u00eda de la pcr, llevando a cabo experimentos in vivo que demostraron la superioridad, en t\u00e9rminos de sensibilidad, del nested-pcr aplicado a muestras de sangre, ya que se obtuvo un 85% de positivos frente al 40% obtenido mediante rt-pcr y southern blot y frente al 5% de positivos obtenidos mediante aislamiento en cultivo celular. por otro lado, se evalu\u00f3 la diversidad gen\u00e9tica de un grupo de 32 aislados de vhsv con distinto origen, tanto
