En este trabajo de tesis, hemos demostrado que ambos receptores junto con el citoesqueleto de actina y prote\u00ednas relacionadas tales como ezrina, moesina, alpha-actinina, vinculina y vasp participan en la formaci\u00f3n de una estructura tridimensional (estructura de anclaje endotelial) que emerge de la cara apical de las c\u00e9lulas endoteliales activadas para envolver parcialmente y adherir firmemente a los linfocitos, impidiendo su desuni\u00f3n en condiciones hemodin\u00e1micas fisiol\u00f3gicas. La formaci\u00f3n y el mantenimiento de esta estructura con forma de copa est\u00e1n regulados por rho\/rock y el fosfoinos\u00edtido pip2. Adem\u00e1s, las tetraspaninas cd9 y cd151 se asocian lateralmente con icam-1 y vcam-1 en la membrana plasm\u00e1tica, y colocalizan con los receptores de adhesi\u00f3n en las estructuras endoteliales de anclaje. Haciendo uso de rna de interferencia para tetraspaninas y de p\u00e9ptidos bloqueantes espec\u00edficos de cd9, hemos demostrado que la inserci\u00f3n de icam-1 y vcam-1 en microdominios de tetraspaninas es crucial para el adecuado funcionamiento adhesivo de estos receptores endoteliales. Por otra parte, icam-1 y vcam-1 son capaces de co-agruparse en las estructuras de anclaje en ausencia de uni\u00f3n a ligando, anclaje a actina y formaci\u00f3n de heterod\u00edmeros. Este fen\u00f3meno se puede explicar por las interacciones espec\u00edficas que tienen lugar dentro de los microdominios de tetraspaninas (icam-1\/cd9, vcam-1\/cd151, cd9\/cd151) y que han sido caracterizadas haciendo uso de t\u00e9cnicas de microscop\u00eda anal\u00edtica (flim-fret). El an\u00e1lisis de las propiedades biof\u00edsicas de las tetraspaninas y los receptores de adhesi\u00f3n mediante espectroscop\u00eda de correlaci\u00f3n de fluorescencia demuestra la existencia de estos microdominios organizados que contienen cd9, cd151, icam-1 y vcam-1 en la membrana plasm\u00e1tica apical de c\u00e9lulas endoteliales vivas, con prop<\/p>\n<p>&nbsp;<\/p>\n<h3>Datos
