P53 es un regulador maestro que responde a distintos da\u00f1os celulares. Entre ellos, el da\u00f1o en el adn y la se\u00f1alizaci\u00f3n oncog\u00e9nica son omnipresentes en c\u00e1ncer y activan p53 a trav\u00e9s de v\u00edas moleculares distintas. El da\u00f1o en el adn se se\u00f1aliza hacia p53 mediante una cascada de quinasas que incluye a atm, atr, chk1 y chk2; mientras que la se\u00f1alizaci\u00f3n oncog\u00e9nica activa arf, que a su vez media la activaci\u00f3n de p53. Una vez activada, p53 ejecuta un programa transcripcional que desemboca en distintas respuestas celulares, dependiendo del tipo celular y del grado y duraci\u00f3n del est\u00edmulo, entre otros factores. Esta activaci\u00f3n puede provocar parada del ciclo celular, fundamentalmente por la transactivaci\u00f3n de p21, o apoptosis, por la transactivaci\u00f3n de otros genes diana como puma, noxa o bax. Sin embargo, la importancia relativa de cada uno de estos activadores y respuestas celulares en la supresi\u00f3n tumoral mediada por p53 se desconoce, en parte por la falta de modelos y herramientas adecuadas para investigar las respuestas mediadas por p53 in vivo.  los ratones super p53, desarrollados en nuestro laboratorio, son ratones transg\u00e9nicos que portan una copia gen\u00f3mica adicional del gen p53, con regulaci\u00f3n end\u00f3gena y expresi\u00f3n normal. Por lo tanto, los ratones super p53 poseen tres copias del gen p53, y por ende presentan una actividad de p53 aumentada y una marcada protecci\u00f3n antitumoral. Hemos cruzado los ratones super p53 con ratones p21-nulo y evaluamos gen\u00e9ticamente la dependencia de p21 en la supresi\u00f3n tumoral mediada por p53.
