Estudios epidemiol\u00f3gicos y precl\u00ednicos indican que la 1 ,25-dihidroxivitamina d3 (1,25(oh)2d3), el metabolito m\u00e1s activo de la vitamina d en nuestro organismo, tiene efectos protectores frente al crc. Resultados previos de nuestro laboratorio muestran que la 1,25(oh)2d3 y algunos de sus an\u00e1logos inhiben la proliferaci\u00f3n y promueven la diferenciaci\u00f3n epitelial de las c\u00e9lulas sw480-adh de c\u00e1ncer de colon humano. Estas acciones se basan, principalmente, en la inducci\u00f3n transcripcional del gen cdh1\/e-cadherina y en el bloqueo de la ruta wnt\/ -catenina, muy frecuentemente alterada en esta neoplasia. La mayor\u00eda, si no todas, las acciones de la 1,25(oh)2d3 est\u00e1n mediadas por el receptor de vitamina d (vdr), un factor de transcripci\u00f3n de la superfamilia de los receptores nucleares. %&#038;\/Los resultados presentados en esta memoria muestran que la 1,25(oh)2d3 regula genes implicados en distintas funciones celulares, siendo el patr\u00f3n de expresi\u00f3n g\u00e9nica inducido dependiente de las caracter\u00edsticas de cada l\u00ednea celular de c\u00e1ncer de colon estudiada. La 1,25(oh)2d3 cambia los niveles de expresi\u00f3n de numerosos genes en las c\u00e9lulas sw480-adh, de modo acorde con su efecto antitumoral. %&#038;\/El estudio del mecanismo de acci\u00f3n de la 1,25(oh)2d3 ha revelado que \u00e9sta induce la activaci\u00f3n transitoria de la v\u00eda rhoa-rho associated coiled kinase (rock). Esta activaci\u00f3n es precedida por un aumento de la concentraci\u00f3n citos\u00f3lica de ca2+, que requiere vdr pero es independiente de expresi\u00f3n g\u00e9nica. Posteriormente, la 1,25(oh)2d3 activa la prote\u00edna quinasa activada por mit\u00f3genos p38 (p38mapk) y la prote\u00edna quinasa activada por mit\u00f3genos y estr\u00e9s (msk)1. La activaci\u00f3n de rhoa-rock y p38mapk-msk1 es necesaria para la inducci\u00f3n por la 1,25(oh)2d3 de un fenotipo adhesivo epitelial y de cdh1\/e-cadherina, cyp24 y otros genes. Adem\u00e1s, rhoa-rock y msk1 est\u00e1n implicadas en la inhibici\u00f3n de la ruta wnt\/ -catenina y de la proliferaci\u00f3n celular por la 1,25(oh)2d3. Estos hallazgos indican que los efectos de la 1,25(oh)2d3 en c\u00e9lulas humanas de carcinoma de colon dependen de una acci\u00f3n dual de vdr como activador r\u00e1pido de rhoa-rock y p38mapk-msk1 y como factor de transcripci\u00f3n, lo que implica la conexi\u00f3n entre acciones no-gen\u00f3micas y gen\u00f3micas de la 1,25(oh)2d3.
