{"id":61578,"date":"2018-03-09T22:49:25","date_gmt":"2018-03-09T22:49:25","guid":{"rendered":"https:\/\/www.deberes.net\/tesis\/sin-categoria\/estudi-funcional-del-gen-dor\/"},"modified":"2018-03-09T22:49:25","modified_gmt":"2018-03-09T22:49:25","slug":"estudi-funcional-del-gen-dor","status":"publish","type":"post","link":"https:\/\/www.deberes.net\/tesis\/bioquimica\/estudi-funcional-del-gen-dor\/","title":{"rendered":"Estudi funcional del gen dor"},"content":{"rendered":"<h2>Tesis doctoral de <strong> Jordi Duran I Castells <\/strong><\/h2>\n<p>Con el objetivo de identificar genes involucrados en la fisiopatolog\u00eda de la diabetes mellitus de tipo 2 se realizo un estudio de hibridaci\u00f3n substractiva entre ratas zdf (obesas i diab\u00e9ticas) i ratas control.  se identificaron diversos clones que estaban diferentemente expresados en las dos condiciones; uno de ellos correspond\u00eda a dor, que se llam\u00f3 as\u00ed por la siglas en ingles de \u00abdiabestes and obesity regulated\u00bb. El gen dor no hab\u00eda estado descrito previamente, por lo que el principal objetivo de esta tesis ha sido identificar i caracterizar la funci\u00f3n celular de dor. As\u00ed en el an\u00e1lisis de la seq\u00ed\u00bcencia de la prote\u00edna se identificaron varios elementos que indicaban que dor pod\u00eda ser un coactivador de receptores nucleares de hormonas. Para comprobar esta posibilidad se realizaron ensayos con vectores reporter que demostraron que dor coactiva diversos receptores nucleares, entre ellos el receptor de hormonas tiroideas, el receptor de glucocorticoides ippar. Estudios con una prote\u00edna de fusi\u00f3n de la prote\u00edna dor con el dominio de uni\u00f3n de dna del factor de transcripci\u00f3n de levadura gal4 demostraron que la prote\u00edna dor es capaz de activar por si misma la transcripci\u00f3n. Asi, dor posee dos dominios de activaci\u00f3n de la transcripci\u00f3n independientes. En los dos dominios encontramos un grupo de residuos hidrof\u00f3bicos que son importantes para la activaci\u00f3n de la transcripci\u00f3n, ya que su mutaci\u00f3n disminuye la capacidad activadora de la fusi\u00f3n gal4-dor. Estudios de microscopia confocal demostraron que la prote\u00edna dor se localiza principalmente en el n\u00facleo en c\u00e9lulas proliferativas. Esta localizaci\u00f3n nuclear viene mediada por una regi\u00f3n c-terminal de la prote\u00edna. Dor presenta adem\u00e1s una se\u00f1al de exporte nuclear, que es responsable de la salida del n\u00facleo de dor en determinadas circunstancias. La prote\u00edna podria estar regulada por diversas modificaciones post-trafuccionales que influir\u00edan en su localizaci\u00f3n subcelular, as\u00ed como en su actividad y su vida
