La v\u00eda de wnt permite controlar los niveles de beta-catenina citoplasm\u00e1ticos mediante la degradaci\u00f3n proteos\u00f3mica. El acumulo de beta-catenina en el citoplasma facilita la traslocaci\u00f3n nuclear de beta-catenina con la consiguiente activaci\u00f3n de los miembros de la familia de factores de transcripci\u00f3n t-cell factor\/lymphoid enhancer factor (tcf\/lef) y la expresi\u00f3n de genes diana como c-myc, cyclin-d1 y tcf-1, favoreciendo el crecimiento y diferenciaci\u00f3n celular. la activaci\u00f3n aberrante de la v\u00eda de wnt se ha asociado al c\u00e1ncer. Est\u00e1n descritas mutaciones que son directamente oncog\u00e9nicas como las mutaciones en el gen de la adenomatous poliposis coli (apc) y tambi\u00e9n aunque menos frecuentes las de beta-catenina (ctnnb1) y axin en c\u00e1ncer colorectal. No obstante, est\u00e1 menos claro cual es el papel de los ligandos extracelulares as\u00ed como los antagonistas de \u00e9sta v\u00eda de activaci\u00f3n en el proceso de carcinog\u00e9nesis. Recientemente, en c\u00e1ncer colorectal se ha asociado la hipermetilaci\u00f3n del promotor de un antagonista de la v\u00eda de wnt frizzled (sfrp) como causa directa de silenciaci\u00f3n prot\u00e9ica. M\u00e1s interesante fue la observaci\u00f3n de que la re-expresi\u00f3n de sfrp en l\u00edneas celulares de c\u00e1ncer colorectal aten\u00faa la se\u00f1alizaci\u00f3n de esta v\u00eda de activaci\u00f3n incluso en presencia de mutaciones intr\u00ednsecas. la presente tesis doctoral pretende evaluar el papel de los ligandos extracelulares de wnt en l\u00edneas celulares de c\u00e1ncer colorectal portadoras de mutaciones as\u00ed como la caracterizaci\u00f3n e identificaci\u00f3n del \u00e1rea promotora del factor inhibidor de la v\u00eda de wnt (wif)-1.
