En determinadas ocasiones es posible asignar funciones por comparaci\u00f3n de secuencia con otras prote\u00ednas conocidas. Sin embargo, en el caso de la enzima que nos ocupa, llamada maleilacetoacetato isomerasa (maai), la comparaci\u00f3n de secuencias puede inducirnos a errores respecto a la funci\u00f3n de la prote\u00edna.  esta enzima fue clonada, in silico, en 1997 y clasificada como una nueva clase de glutati\u00f3n transferasa (gstz) por comparaci\u00f3n de secuencias, a pesar de que los autores s\u00f3lo fueron capaces de demostrar una m\u00ednima actividad gst. Independientemente, en nuestro grupo clonamos la enzima como la aut\u00e9ntica maai, esto es, la enzima perteneciente a la ruta catab\u00f3lica de fenilalanina y tirosina que convierte el maleilacetoacetato (maa) en su is\u00f3mero, el fumarilacetoacetato (faa).  Adem\u00e1s de estas actividades maai y gst, se han encontrado otras actividades enzim\u00e1ticas asociadas a esta prote\u00edna. Una de ellas, es una actividad que realiza la deshalogenaci\u00f3n oxidativa del \u00e1cido dicloroac\u00e9tico (dca), que es convertido a glioxilato. El \u00e1cido dicloroac\u00e9tico (propuesto como cancer\u00edgeno) se origina en la cloraci\u00f3n del agua y se utiliza como terapia en la acidosis l\u00e1ctica. La otra actividad asociada a esta prote\u00edna es una actividad peroxid\u00e1sica, que es t\u00edpica de algunas gst. existen dos organismos modelo deficientes en maai (un microorganismo y un rat\u00f3n) para su estudio, y hemos encontrado otro substrato para la enzima, el \u00e1cido mal\u00e9ico. En mam\u00edferos no se ha descrito ninguna actividad enzim\u00e1tica que degrade el \u00e1cido mal\u00e9ico. Solamente hemos encontrado algunas referencias de los a\u00f1os 50 y 60, en que se describe la observaci\u00f3n de que rodajas de ri\u00f1\u00f3n de conejo pueden convertir el maleato en fumarato y\/o malato. En microorganismos si se han caracterizado dos enzimas que convierten el maleato a fumarato y a malato.
